Objective: To establish the acute rejection model of orthotopic liver transplantation in inbred rat, then use PPARγ ligand 15d-PGJ2 in vivo. The aim is to judge if secretion of interleukin-2 is blocked, activation is inhibited and apoptosis is induced of T lymphocytes by 15d-PGJ2 in recipient rat, the last investigate the effects of PPARγ ligand on preventing acute rejection and inducing immune tolerance after liver transplantation in rat.Methods: Surgical skill were trained by using closed colony rat SD and Wistar, following DA to Lewis orthotopic liver transplantation acute rejection models were fulfilled with the modified two-cuffed technique. These inbred rats were divided into 3 groups: group A, the immune tolerance group(Lewis-to-Lewis,n=8), no any intervention; group B, control group(DA-to-Lewis, n=10),1ml NS was injected by i.p.; group C, test group (DA-to-Lewis ,n=10), 15d-PGJ2 200ug·kg-1·d-1 was injected by i.p.(from 1d before operation to 14d after transplantation in group B and C). The clinic symptoms and signs were observed on 5d, 7d after operation. Recipient survival rate, histopathological characteristics of liver tissue were viewed . Plasma levels of TBIL, ALT, AST, ALP and ALB were measured. The expressions of PPARγmRNA in liver tissue were examined by Real-Time fluorescent quantitation RT-PCR.
