Objective: The objectives of this study are twofold. First, we planned to investgate the characteristics PLGA sustained release microspheres (Ms) incorporating Rifampin (RFP) through release in vitro and in the local position of para-vertebra, Second we built spinal tuberculose model by inserting standard tuberculose strain H37Rv into the lumbar spine in rabbits, in order to make an experimental basis for the other upcoming experiments.Methods: (1) RFP-PLGA-Ms using optimized O/W emulsion volatising method and stored by the routine freeze-dried. (2) The Ms electron scan microscope was used to observe the morphology of Ms and determine the particle size distribution of Ms with particle size analyse apparatus. (3) High Performance Liquid Chromatograph (HPLC) was used to construct the standard regression equation curve of RFP. This helped determine the rates of drug loading rate and embedding. (4) PBS solution was used as medium to study the property of RFP-PLGA-Ms release in vitro. (5) RFP-PLGA-Ms was inserted to para-vertebrato study the behavior of Ms about release in vivo.
