Construction And Expression Of A Single-chain Fv Against Bovine Bone Morphogenetic Protein

As an osteocarcinoma-associated antigen,bone morphogenetic pro tein(BMP) is of significant in the clinical study of osteocarcinoma.In 1990, a monoclonal antibody against bovine BMP was made in our department.There were some studies of the bBMP-McAb to show that the bBMP-McAb selectively gathered in the place where new bone formed and osteosarcoma occured in the patients.Immunohistochemical study of the bBMP-McAb showed that the cartilagesarcoma and osteosarcoma cells contained abundent of BMP.Thus it can be seen that bBMP-McAb has significant potential value in the guided therapy for the osteocarcinoma. Because it would cause immunogenic response when the murine antibodies were applicated in patient directively,the usefulness of murine antibody is limited. In recent years, with the development of genetic engineering technique and the successful reconstruction of the murine antibodies, it offers us a new aproach in treatment.
