Odontoblasts are the most important cells in dental pulp. They keep close relationships with dentinogenesis, and play a critical role in the process of tooth development and pulp repair. With the achievements of modern biology more and more attention has been paid to the molecular mechanism and gene control of odontoblast differentiation. However, it's really hard to take tissue samples of odontoblasts from dental pulp because of their anatomical specialty. Up to now, there are few reports on odontoblast in vitro. In this paper being divided into following 3 parts, we aimed at the differentiation and calcification of dental papilla cells, pulp cells and tooth germs in culture which were closely concerned with odontoblasts in order to better understand the characteristics of odontoblasts.1. The Establishment of experimental models for culturing human dental papilla cells, human dental pulp cells and rat tooth germs in vitroIn the present study, fibroblast cells from human pulp, periodontal ligament and gingiva were cultured on the collagen gel isolated from bovine tendon. The results indicated that the success rates of the pulpal and periodontal tissue with gel treatment were 42.9% and 33.3% respectively, there was significant difference between the treatment groups with collagen gel and the controls without it (P<0.01, P<0.05).The dental papilla is a mesenchymal condensation which plays a very important regulatory role during tooth development. The dental papilla cells in vitro can provide a useful model for studying the characteristics of cells. Successful culture of mouse dental papilla cells as monolayers has been reported, but the cells from human have not been present.
