Periodontitis and peri-apical diseases are the main reasons for pathological tooth loss and periodontal ligament fibroblast(PDLF),the main portion of and the most important cell class in periodontum, lays the foundation for periodontal repair and regeneration. While gingival fibroblast(GF),also the main fibroblast component in gingival tissue adjacent to periodontal ligament, shares many similarities with PDLF in many aspects, namely morphology, growth rate and so on,but except in that of functional characterastics. Studies have shown that when GFs were allowed to come into contact with previously exposed root surface, root resorption and periodontal pocket reformation will occur. On the contray, when PDLFs were selectively allowed to come into contact with the root surface, regeneration of a functional periodontum can be observed historically. In this study, we used a recently developed modified gene cloning technique that is established on the basis of PCR and subtractive hybridization technologies, to study the genes, especially new genes that are differentially expressed in cultured primary human PDLFs in comparasion with GFs,so as to unvail the gene expression style of PDLF and thus to underlay the molecular machenism of periodontal regeneration.Additionally, the cloning of...
