The network theory of the immune system regarded as one of the most eminent progresses in the development of modern immunology has been widely accepted and well modified at present. The conception of "internal image" anti-idiotypic(AId) antibody, which can mimic foreign antigenicity, is still the most important one in the theory, and is of theoretical and practical significance not only in immune response regulations, but also in developments of infectious diseases vaccines, pathogenesis of autoimmune diseases, immunotherapy for tumors, and various receptors research works as well. In fact, many receptors have been identified by AId method.Japanese encephalitis(JE) is an acute zoonosis caused by Japanese encephalitis virus(JEV), a member of flaviviridae, with clinical characteristics of high fever, convulsion, conscious disturbance and other meninx-stimulating symptoms. It mainly invades children with case-fatality rate at about 10%, and leaves various sequelas to about 15% survivors. As planned immunization, early diagnosis and clinical treatments improve, JE's morbidity and fatality rates have declined. Yet, its precise pathogenesis is not clearly defined. On account of the role of virus receptors played in viral infection and pathogenesis, it is definitely necessary to deepen the research work on JEV receptor(JEVR). Fortunately, internal image AId method just provides us such a good way to study JEVR that it is possible, by this method, to generate antireceptor antibodies without using the receptor as the immunogen, thus better solving the problems of virus or viral protein(as ligand) purification and bioactivity.In this report, we used anti-JEV antibodies 2H4 and 2F2 with high neutralizing titers to immunize BALB/c mice with large doses by both traditional Freund's adjuvant immunization method and modified nitrocellulose(NC) membrane subcutaneous embedding immunization method.
