Bone morphogenetic proteins (BMPs) are polypeptides that induce ectopic bone formation in standard rat in vivo assay systems. Previous studies have demonstrated the clinical utility of these proteins in fracture healing, spinal fusion and prosthetic joint stabilization. However, the ideal approach to deliver BMPs properly remains unknown. Gene therapy to deliver BMPs offers several theoretical advantages over implantation of a recombinant BMP protein, including persistent BMP delivery and eliminating the need for a foreign body carrier. This study was performed to determine whether fibroblasts can be used to express BMP-2 in vitro and promote endochondral bone formation in vivo by transfer of BMP-2 gene into fibroblasts.1. Construction of recombined eukaryotic expression vector pcDNA3-hBMP2We used gene recombination technology to directly clone the cDNA of hBMP2 into eukaryotic expression vector pcDNA3 and formed a recombined...
