Objective: To observe the therapeutical effect of a complex prescription of Shenxiaokang on DN rats and discuss the mechanism of action in the treatment of DM, then provide experimental data for clinical practice.Methods: The model rats male Wistar were established with multiple injections of low dose STZ and high calorie diet. Kinds of examination methods were applied, including radiommunoassay, biochemistry, light microscope, electron microscrope, immunohistochemical , Affymetrix Co.US. Rat 2302.0 gene expression profiling chip, and real time quantitative reverse transcription polymerase chain reaction technique to observe the effect of Shenxiaokang on indicators such as body weight, kidney weight, kidney weight index,blood glucose, insulin, glyxosylated hemoglobin, total cholesterol, triglyceride, high ednsity lipoprotein, Serum Creatinine, Blood urea nitrogen, Albumin, beta2-microglobulin, Angetension-II, Growth Hormone/Insulin-like growth factors-1, Tumor necrosis facto-Alpha, Interleukin -2, and patho-morphous of kidney, especially gene expression profiling and CXCL10 mRNA and CCL5mRNA gene.Results:(1)The models of DN rats have been cloned successfully.(2) Shenxiaokang has good effects on decreasing FBG, HBAlc, TC, TG, Scr, BUN, Alb,β2-MG and increasing the level of Ins,HDL-c.(3)AngII ,GH/IGF-1,TNF-α, IL-2,NF-κB have important action in DN. Shenxiaokang has good effects on regulating Ang II,GH/IGF-1,TNF-α,IL-2,NF-κB. These indicate that Shenxiaokang has many targets of pharmacological action, it can prevente and treate DN.(4)Shenxiaokang can restore renal glomerulus and renal tubules microstructure pathology damage, protect kidney function and delay renal failure.(5) With gene expression profiling chip technique to study DN, results show that,①By analyzing gene chip,we found that there were 73 genes expression up-regulation and 191 genes expression down-regulation in the DN model group rat kidneys.②There were 168 genes expression up-regulation and 17 genes expression down-regulation in the Shenxiaokang treatment group rat kidneys.③There were 65 genes expression tended to be equivalent. In these genes there were 11 genes expression up-regulation in the DN model group rat kidneys and down-regulation in the Shenxiaokang treatment group rat kidneys . There were 54 genes expression down-regulation in the DN model group rat kidneys and up-regulation in the Shenxiaokang treatment group rat kidneys .(6) With real time quantitative reverse transcription polymerase chain reaction technique to study DN, results show that, the CXCL10 mRNA and CCL5 mRNA genes were up-regulation in the DN model group rat kidneys and down-regulation in the Shenxiaokang treatment group rat kidneys.Conclusions: The results above display that ,Shenxiaokang has good repaired effect on damaged kidney tissue. Shenxiaokang can protect kidney function, Shenxiaokang has effect on many genes expression , especially the CXCL10 mRNA and CCL5 mRNA genes .Tangxinkang can prevent and treat diabetes and related complication through multiple pathways, tache,target.and direct our clinical practice. DN molecular mechanisms relation to genes expression, including the CXCL10 mRNA and CCL5 mRNA genes,that can leat to worsen DN.
