Purpose We have used realtime RT-PCR technique to compare differentiallyexpressed hPOT1 genes among different grades' glioma and normal brain tissue inhuman and explored the relativity between hPOT1 expression and tumor grade. Itmight be helpful to understand the molecular mechanism of glioma formation and setnew standard of tumor grade. We want to know if hPOT1 can be an index todetermine glioma biology. Our research might be a theorical basis to clinicalengagement of more sensitive and more differential molecμfar tumor symbols. Wehave used flow cytometer to determin proliferation index of C6 and TJ905 gliomacloned strain processed by gossypol or 3'-azide-3'-deoxythymidine(AZT) andinvestigated initially the influence of telomerase inhibitor to glioma.Materials and methods We collected 53 glioma specimens excised in the surgeryand 8 "normal" brain tissues. All specimens were confirmed by pathological diagnosis.We measured hPOT1 expression by realtime RT-PCR technique, at that time ACTBgene expression was an endogenous control. We have used flow cytometer todetermin proliferation index of C6 and TJ905 glioma cloned strain processed by3'-azide-3'-deoxythymidine(AZT) or gossypol.Results We made data logarithmic transformation and then did homogeneity ofvariance estimation and LSD(Least-Significant Difference) estimation. We foundhPOT1 mRNA expression in the glioma tissue is obviously higher than it in thenormal brain tissue. hPOT1 mRNA expression in theⅢ/Ⅳgrade glioma tissue ishigher than it in theⅠ/Ⅱgrade. There is no obvious significance (P<0.05). There is noobvious correlation betweenⅢ/Ⅳgrade andⅠ/Ⅱgrade. The proliferation index of C6and TJ905 glioma cloned strain processed by 3'-azide-3'-deoxythymidine(AZT) orgossypol has degraded. Conclusion These data suggest that Pot1 may play an important role in regμlationof telomere length, telomere maintaining and inhibition of Pot1 may induce telomeredysfunction. Moreover, changes in POT1 expression levels may be associated withglioma carcinogenesis and glioma progression, hPOT1 mRNA expression level issignificantly higher in glioma tumor than in normal brain tissue. We consider thatPOT1 expression level has significant correlation with tumor. Telomerase inhibitormay be new chemotherapeutics for glioma. Gossypol may be a new kind of telomeraseinhibitor.
