Academic And Experimental Studies On Anty-Epilepsy Of Methods For Invigorating QI, Dispelling Phlegm And Tranquilizing The Mind

Objective: The purpose of this study was to present new opinion on the pathogeny and pathology of Epilepsy and establish the therapeutic method for preventing and treating Epilepsy.Observing the anty-EP effects of AAP(Drugs for Invigorating QI,added with Dispelling phlegm and Tranquilizing the mind) on mice induced by PTZ,and investigating the effects of AAP on apoptosis,signal conduction of hipocampus neuron cells with Epilepsy rats;to explore the function and mechanism of Invigorating QI,added with Dispelling phlegm and Tranquilizing the mind for preventing and treating EP,which may offer theoretical support and experimental basis for new type of drugs and clinical application.Methods: Searching about studies of classical and present age on Epilepsy;Observeing the escape latency of searching the platform by means of water maze test and the changes of the latent period,time span,intensity of PTZ induced mice,to explore the effects of AAP on the ability of learning,memory and the behaviors scales with epilepsy mice;Detecting the expression levels of caspase-3,PKCα,and mGluR1 at hippocampus neurons with immunohistochemical techniques and TUNEL,to investigate the effects of AAP on apoptosis and signal conduct of hipocampus neurons.Results: In TCM theory,the pathogeny of epilepsy includes congenital factor,internal injury due to emotional disorder,brain trauma and the secondary affection et al.The pathology is hypofunction of viscera,dysfunction of qi,accumulation of phlegm-dampness in the mind,all of that causing abnormity of mind and mental retardation.The main pathology is phlegm.Our results suggest that Epilepsy is a symptom complex-deficiency of Qi as BEN(primary) and involving phlegm as BIAO(secondary).Deficience of spleen is apt to generate dampness and accumulate phlegm,which is the key of it.Treatment focusing on relieving both the secondary and primary symptoms.The methods of Invigorating QI,added with dispelling phlegm and Tranquilizing the mind should be taken.The AAP can tranquilizing the mind and promoting intelligence by invigorating qi and enriching blood,resolve phlegm to open orifice.The results show that the escape latency of searching the platform were shortened and the mistakes made were less;the latent period were prolonged and the time span were shortened with AAP;and the average optical density of caspase-3,mGLuR1 and PKCαof AAP group were obviously decreased.Conclusions: It is concluded that Invigorating QI,added with Dispelling phlegm and Tranquilizing the mind is one of the most important measures to intervene with Epilepsy.Deficiency of spleen as BEN(primary),phlegm as BIAO(secondary),and it carries out method of Invigorating QI,Dispelling Phlegm and Tranquilizing the Mind.The therapeutic principle is strengthening healthy qi to eliminate pathogenic factors;and the therapeutic method is strengthening the spleen for dispelling phlegm.The AAP can improve the capability of learning and memory of epilepsy mice induced by PTZ effectively. AAP can protect neuron effectively by decreasing the expression of PKCαand mGluRl,also can decrease the expression of caspase-3,inhibiting apoptosis of haipocampus neuron cells.we conclude that AAP can invigorating qi and enriching blood,resolving phlegm to open orifice,tranquilizing the mind and promoting intelligence.
