Objective:1. To master the techniques of three-dimensional chondrocyte culture in vitro,the establishment of the model of auto-articular cartilage full—thickness defect and autrograft of the cultured chondrocytes plus auto-periost covering the defected aera.2. To explore the effects of repair of articular cartilage full—thickness defect by chondrocytes cultured in alginate in rabbits autograph.3. To provide a basic study for the clinical treatment of the articular cartilage disease.Methods:1. The source of the auto-chondrocyte the chondrocytes are come from the condyles of femur of four weeks newsland rabbits under abdominal anthesia, digested by machine and enzyme,and then, make them into cell suspension.2. The transplant of the auto-chondrocyte the three-dimensional cultured chondrocytes are digested into single chondrocyte,and then,the suspension of the chondrocyte were injected into the defect area of the articular cartilage which are covered by the periost of the proximal wall of the auto-tibia.3. The groups of the experiment the 24 rabbits were divided into four groups: the auto-chondrocyte transplant group, the auto-control group(with the covering of the periost),the variant control group(with the covering of the periost) and the variant control group(without the covering of the periost),6 rabbits each group.4. The design of the experimental model of the defected articular cartilage two cylindrical,full-thick articular defects,5.0mm in diameter, were produced in the left knee joints of 24 rabbits.they were divided into four groups according to the design of the experiment.5. Evaluation of the histology: the animals were killed respectively in 4,8,12,16 weeks, the distal femur were cutted.The macroscopic examination, H-E stain were performed.the collagen II was observed immunologically.Results1. The chondrocytes digested by machine and enzyme were successfully cultured in monolayer and three dimensional culture system. The three dimensional culture model were successfully established 2. The model of the auto-articular cartilage full—thickness defect and the technique of the autrograft of the cultured chondrocytes plus auto-periost covering the defected aera were successfully performed.3. The repairing effect of the three-dimensional cultured chondrocytes is satisfactory in short time, and the results of the evaluation of the histology is better than the contrast groups.Conclusions1. To gain a large amount of good phenotype chondrocytes in a short period is possible in three-dimensional culture system.2. The repairing effects of the autograft of the chondrocytes to the articular cartilage full—thickness defect area is good in short time.
