Functional malocclusion is common, and its treatment method is functional orthopedics. The morphological and functional remodeling of masticatory muscles happens initially in a new mechanical environment, which is the key of functional orthopedics and plays an important role in the treatment results and retention. The shape and function of masticatory muscles are tied up with shape and function of myoblast. So studying the shape and function of myoblast at the cell level or the molecular level is the basis for us to understand the mechanism of orthopedics under stress environment by cell biomechanic methods in vivo.It is difficult to reflect the condition of extracellular matrix (ECM) of The biomechanics study of cell monolayer in vitro. So we have built the model of 3D about myoblast and the poly lactide-co-glycolide (PLGA) scaffold. Then, we study the correlation factors of cytoskeleton and MGF of cultured SD rat myoblast on the PLGA scaffold induced by cyclic strain.The results show as following:(1) The PLGA scaffold, whose the interval porosity is 85% and the pore size is 88~100μm, is a satisfactory scaffold of tissue engineering of myoblast.(2) The myoblast can express integrinβ1,α-actin and mechano-growth factor (MGF). The tendency of the express are not different between the myoblast of 3D culture and those of 2D culture.(3) The express of integrinβ1,α-actin and MGF are closely correlating with loading time prolonged.(4) There are some differents of express of inegrinβ1,α-actin and MGF between the myoblast of 3D culture and those of 2D culture.Based on these findigs, at the physiologica strain, the express of integrinβ1,α-actin and MGF are closely correlating with loading time prolonged. some differents of function express of the myoblast are existing between the myoblast of 3D culture and those of 2D culture.The results are useful for us understanding some phenomenon, such as the wearing time of the appliance of orthopedics according with the time of the top of the express of integrinβ1 and MGF of myoblast.
