Objective: To evaluate the effect of bone mesenchymal stem cell transferred by hBMP-7 gene to repair rabbits degenerative disc. Methods: BMSC was infected by recombinant adenovirus carrying hBMP-7 gene and the expression of the gene was determined by immunocytochemistry. The rabbit model of degenerative intervertebral disc was established by nucleus aspiration. BMSC and gene transferred were transplanted into the degenerative disc respective.Immunohistochemistry, RT-PCR and Western- blot was used to detect hBMP-7 expression. The effect of repair the degenerative disc was determined by MRI, immunohistochemical and biochemical analysis. Results: Immunohistochemistry,RT-PCR and western- blot demonstrated the expression of hBMP-7 in the groups of gene transferred BMSC. The data which included the height and T2 value of the disc, the content of proteoglycan and Typeâ…¡collagen, have significant difference among the three groups. Conclusions: HBMP-7 gene transferred BMSC may be used to repair the degeneration disc. Further study are needed to define the procedure of BMSC differentiation in vivo.
