Now laboratory animals have become one of most elementary research tool of genetics, disease and pharmaceutical exploitation et al research field. For sharing integrating of biomedical information more conveniently, synchronously considering the rights of animal survival and welfare, digitization of structure and function information on small animals has become research topic of many scientific research institutions. Digital anatomy atlas has drawn attention of more and more scientists because of its exact, intuitionistic and multi-formal graphics representation and calculability of anatomy models.Through analyzing and comparing different capturing method of 3D section anatomy images, cryosection imaging system for small animal study was developed based on cryosection technology. In this paper, SD rat anatomy atlas was build based on cryosection imaging system for small animal study, and the constructing method, technique scheme of section anatomy atlas, technique parameters selecting, possible problem and its solving answer were discussed systemically. And constructing SD rat digital atlas based on SD rat anatomy atlas, constructing technique scheme and relative technology were discussed.Main results of the study are as follows:1. Based on cryosection technology, cryosection imaging system for small animal study was developed. System parameters are as follows: Minimum feed of milling machine was 0.001 mm; repeatability of positioning was 0.01/300 mm. The features of the system are: suiting gross specimen imaging, high-resolution, destructive imaging, having intrinsic tissue texture and color of specimen. Cryosection-milling imaging system, which can be as a perfect tool to obtain precise structural information of small animal, offers a new efficient method for small animal study.2. In this paper, the cryosection-milling imaging process of a SD rat was described, and the SD rat atlas was constructed based on cryosection-milling imaging system. The voxels size of SD rat atlas was 20μm×20μm×20μm, the cryosection images were shot with the resolution of 4,600×2,580×24 bit and in total of 9475 slices and 314.68 GB. And Vascular Perfusion SD rat atlas was constructed based on cryosection-milling imaging system. The voxels size of SD rat atlas was 20μm×20μm×20μm, the cryosection images were shot with the resolution of 4,917×3,446×24 bit and in total of 1646 slices and 77.9 GB.3. Through discussing technique scheme and constructing technique of digital atlas, SD rat digital anatomy atlas was constructed to obtain high-precision 3D SD rat anatomy models and artery system model, based on SD rat atlas and Vascular Perfusion SD rat atlas. These models have extensive education and practice application value.4."Visual Rat Atlas"and"Visual Rat Explorer"were programmed to separately show section images, anatomy map of different sections and 3D SD rat models. These software are convenient, applied bio-research tool, through which user can analyze and compute biomedical data.Research proves cryosection imaging system for small animal study provide a effective means for studying structure information of small animal. Digital anatomy atlas can be a framework to integrate more biological information for studying different levels biological information, and provide applied models for small animal study and in biomedical research.
