Hematopoietic growth factors are an important part of cytokines, which show an important role in the mechanism of blood cells building and regulation. Hematopoietic growth factors mainly consist of Erythropoietin (Epo), Granulocyte colony-stimulating factor (G-CSF), Granulocyte-Macrophage colony-stimulating factor (GM-CSF), multipotent colony-stimulating factor or Interlukin 3 (Multi-CSF or IL-3), Macrophage colony-stimulating factor (M-CSF), Stem cell factor (SCF), Thrombopoietin (Tpo). Among them, SCF is different, which can act directly on early hematopoietic stem cells and primitive hematopoietic progenitor cells and induce the survival, proliferation and differentiation of these cells. SCF shows strong synergistic roles with other cytokines and remarkably reduces the quantities of other cytokines although it has low effect alone. GM-CSF is a regulator of the intermediate stage of hematopoiesis. It supports the expansion and growth of both granulocytic and monocytic colony forming units, as well as colonies that contain myeloid, erythroid and megakaryocytic cells and macrophages and neutrophils. Basing on the synergistic characteristics of SCF and other cytokines, we designed and constructed two kinds of new-style fusion proteins: human SCF-GM-CSF. In attempts to maintain or enhance the effects of the two individual molecules, we combine human SCF and human GM-CSF, which has similar and cooperative functions, into one molecule.There are two main parts in this paper: High expression plasmid including human SCF gene was constructed and hSCF was purified and identified. High expression plasmids including human SCF-GM-CSF fusion genes were constructed and fusion proteins were purified and identified.
