Objective: To observe the therapeutic effect of Yangxin containing serum induced hydrogen peroxide in human umbilical vein endothelial cells from oxidative injury model of cell proliferation , differential gene expression and Smad4 mRNA influence the level of molecular biology explore possible mechanisms for objective basis for clinical application.Method:1. The 20 male wistar rats were randomly divided into control group and Yang Xin Tang group, n=10. Yang Xin Tang were given orally Yang Xin Tang, 2 times daily administration, the blank group were given orally with saline, 2 times daily administration, are continuous administration 1 week after the last administration blood.2.Experiment divided into①blank serum;②model group;③Yang Xin Tang group containing 2% serum;④Yang Xin Tang 4% in group containing serum;⑤Yang Xin Tang group containing 6% serum;⑥Yang Xin Tang 8% in group containing serum.3.Hydrogen peroxide in human umbilical vein endothelial cells induced by oxidative damage, similar to atherosclerosis copy Endothelial injury model.4.With inverted microscope , MTT assay Yang Xin Tang serum containing hydrogen peroxide injury of endothelial cell proliferation.5.Using Affymetrix human genome U133 +2.0 Yang Xin Tang microarray containing serum on hydrogen peroxide injury of endothelial cells in different gene expression.6.Using real-time quantitative PCR was used to detect drug-containing serum Yang Xin Tang Hydrogen Peroxide damage to the endothelial cells the expression of Smad4 mRNA.Results:1. From observed under the inverted microscope, 2%, 4%, 6%, 8% Yangxin containing serum can reduce hydrogen peroxide on endothelial cell damage, of which 6% concentration of the best. 2.2%, 4%, 6%, 8% Yangxin containing serum can increase the rate of proliferation, of which 6%, 8% concentration of the best, and the model group, the difference was statistically significant (P <0.01). 6%, 8% Yangxin containing serum between the two groups, the difference was not statistically significant (P> 0.05) .3. On human umbilical vein endothelial cell gene expression profile results showed that: the model group compared with the control group, 132 differentially expressed genes, including 84 upregulated genes, 48 genes were down; Yang Xin Tang difference between group and control group 1645 genes, of which 1 371 upregulated genes,274 genes were down; which the model group compared with the Yang Xin Tang, 35 differentially expressed genes, upregulation of the model group, after control by Yang Xin Tang decreased expression of 23 genes section, the expression of the model group down after the treatment by Yang Xin Tang regulated genes 12. After MAS analysis, its molecular function and biological process negative regulation of cellular proliferation, cell growth-related genes SAMD4; and negative regulation of cell migration-related genes NF1; and angio -genesis-related genes have NF1, SHB so. 4.6% concentration of Yang Xin Tang can lower serum containing hydrogen peroxide injury of endothelial cell expression of Smad4 mRNA.Conclusion:1. Yangxin containing serum can reduce hydrogen peroxide on endothelial cell injury, increased endothelial cell proliferation rate, which may be the prevention of atherosclerosis in one of the mechanisms.2. Yang Xin Tang control serum with hydrogen peroxide injury of endothelial cells in gene expression. These genes and cell proliferation and growth, cell cycle, cell migration, angiogenesis, and so on,which may be the prevention of atherosclerosis in one of the mechanisms.3. Yang Xin Tang lowered serum with hydrogen peroxide injury of Smad4 mRNA expression in endothelial cells, which may be the prevention of atherosclerosis in one of the mechanisms.
