Objective:To study the influence of Chinese medicinal herbs with hot property such as aconite root and zingiberis and herbs with cold property such as coptis chinensis and rhubarb on whole genome expression of rat liver. To analyse the possible mechanism of hot and cold effects from the view of gene expression spectrum and to explore the research method of Chinese medicinal property theory and possible basis on definiting the attribute of hot and cold property.Methods:Fifty male SPF Wistar rats were randomly divided into aconite root, zingiberis, coptis chinensis, rhubarb and control groups, 1ml concentrated solutions of aconite root, zingiberis, coptis chinensis, rhubarb and NS were respectively intragastric administrated to rats in each group once daily for 20 days.20 days later, the rat liver gene expressions in each group were dected by RatRef-12 gene array made by Illumina Company. The differential expression gene was selected and cluster analyaed. A part of genes were selected to test the accuracy of results by PCR.Results:As compared to the control group, in the aconite root group there were 592 strips of gene differential expression, and 48 items of significant gene function mainly involving in 7 GO function subtypes such as metabolic process, catalytic activity, immune reaction, et al., especially for metabolic process. As compared to the control group, in the zingiberis group there were 1189 strips of gene differential expression, and 98 items of significant gene function mainly involving in 8 GO function subtypes such as metabolic process, immune reaction, catalytic activity, combination activity, et al., especially for metabolic process. As compared to the control group, in the aconite root and zingiberis groups there were 337 strips of common gene differential expression, and 60 items of significant gene function mainly involving in 7 GO function subtypes such as metabolic process, immune reaction, catalytic activity, ionic transport, transport activity, stress responder and transcriptional control gene function by turns, especially for metabolic process. As compared to the control group, in the coptis chinensis group there were 399 strips of gene differential expression, and 44 items of significant gene function mainly involving in 7 GO function subtypes such as defence reaction, chemotaxis, metabolic process, immune reaction, et al., especially for defence reaction. As compared to the control group, in the rhubarb group there were 398 strips of gene differential expression and 32 items of significant gene function mainly involving in 6 GO function subtypes such as immune process, metabolic process, defence reaction, catalytic activity, ea al., especially for immune and metabolic process. As compared to the control group, in the coptis chinensis and rhubarb groups there were 141 strips of common gene differential expression, and 13 items of significant gene function mainly involving in 3 GO function subtypes such as ferri ion combination, organic acid metabolism and responding to exterior stimulation gene function by turns.Conclusion:Chinese medicinals affect organism on many targets by different pathways. This study discovered that the herbs with hot property could affect the metabolism process of sugar, lipoid and amino acid and promote the production, storage and utilization of energy by regulating the related gene expression of metabolism and catalytic activity and regulate immune function by controlling the related gene expression of immunization, which maybe the main molecule mechanism of warming yang and dissipating cold and to treat cold pattern of hot and warm medicinals. The herbs with cold property have antinflammatory, antibiosis and controlling apoptosis functions by regulating the gene expressions related to defence reacition, apoptosis and immune response. The herbs with cold property can depress the pathological accentuation and enhance the defence and response function, which maybe the main molecule mechanism of clearing heat and purging fire and to treat the pyrectic pattern.
