Objective:To study the regulation of the mice IFN-y overexpress lentivirus on CXCR4 in the RAW264.7 cells, and the influence of the mice IFN-yoverexpress lentivirus on collagen-induced arthritis animal model.Methods:1.RAW264.7 cells were infected with the mice IFN-yoverexpress lentivirus, observed with fluorescence microscope, measured the density of the IFN-y of the cells supernatant with Elisa and the expression of the CXCR4 with western blot.2. six to eight weeks old male DBA/1 J mice were divided into three groups randomly:without intervention with collagen group(normal group), with intervention with collagen group 1 (also with intervention with negative control lentivirus group), with intervention with collagen group 2 (also with intervention with IFN-yoverexpress lentivirus group). Bovine collagen II (BCII,150ug each mice) and complete Freund's Adjuvant (CFA) were emulsionized thoroughly on the ice, injected intradermally on the tail, then, on the 21 day, 100ug BCII and incomplete Freund's Adjuvant (IFA) were emulsionized thoroughly and also injected intradermally on the tail, at the same time, virus were injected in the caudal vein.During the whole immunologic process,we observed the ankle joints and toe joints, and measured the diameter of the pathological ankle joints. At last, we took the blood plasma and bone tissue of all the mice, tested the IFN-y of the plasma and stained the bone tissue section.Results:a) The mice IFN-yoverexpress lentivirus could infected RAW264.7 cells, and made the expression of the IFN-y increased more significantly than noninfected cells or infected with negative infection.b) The mice IFN-yoverexpress lentivirus could infected RAW264.7 cells, and made the expression of the CXCR4 decreased more significantly than noninfected cells or infected with negative infection.c) Collagen induced arthritis animal model were established succeedly, the incidence of the inflamentary joints of the group which was infected with IFN-yoverexpress lentivirus was lower than negative group.d) The density of the plasma IFN-y of the group which was infected with IFN-yoverexpress lentivirus was lower than normal group and negative group.e) The pathology changes of the group which was infected with IFN-yoverexpress lentivirus was lower than normal group and negative group.Conclusion:IFN-yoverexpress lentivirus could infect both RAW264.7 and collagen induced arthritis animal model effectively; IFN-ycould increase the desity of the IFN-y significantly and decrease the expression of the CXCR4 significantly; IFN-yalso could lighten the pathology of the collagen induced arthritis animal models. ObjectiveTo investigate the prevalence of the osteoporosis in Wuhan and look for the scientific basis for the prevention and treatment of osteoporosis.MethodsBone mineral density(BMD) assessment of right calcaneal region (11559 citizens who had medical examination in Hubei Province Xinhua hospital from the year 2004 to 2008, including 6434 men and 5125women, aged from 20 to 99) were measured with dual energy X-ray absorptiometer (DXA). Analysis the BMD and the prevalence of osteopenia or osteoporosis of the subjects, who were divided into groups according to different years, age and sex.ResultsMale and female had significant differences of the BMD,no matter in which year or age group, in case of the same bone mass status (P<0.05).The prevalence of osteopenia or osteoporosis in the year of 2007and 2008 was lower than 2004(P<0.05).ConclusionsFemales have more liability to get osteoporosis than males. The prevalence of osteopenia or osteoporosis in the year of 2007-2008 was lower than 2004-2006,and increaseed with age, especially for women over 50 years old and men over 60 years old.
