Objective:To observe the effect on TNF-α, IL-6, TGF-β1 in ALI rat model, to approach the mechanism of Tongfu Decoction anti-ALI, and to provide experimental base on the lung and the large intestine being interior-exteriorly related theory.Method:180 Wistar rats were divided into 6 groups randomly:the normal control group, the model group, hexadecadrol group, Tongfu Decoction low-dose group, medium-dose group, and high-dose group,30 rats in each group. Copying ALI model by peritoneal injecting LPS, giving respectively commensurate different dose Tongfu Decoction intragastric administration and Dexamethasone intraperitoneal injection half an hour later, and executing rats in the 1,2,6 hour after medication respectively,10 rats each time in each group. We observed lung and intestine histomorphology changes, detected TNF-α, IL-6 content in serum lung and intestine, TGF-(β1 content in lung and intestine by ELISA method synchronously, NF-κB expression by immunohistochemisty and RT-PCR.Result:1. Lung and intestine in the model group showed dark red, turgent, bleeding by naked eye, but other treatment groups showed lighter than it. Lung and intestine in the model group showed inflammatory cell infiltrating, edema, hyperemia, disorganization in light microscope, but other treatment groups showed lighter.2. TNF-αin model serum, lung and intestine reached crest value; and IL-6 in model serum, lung and intestine; TGF-β1 in model lung and intestine increased as time extending in first 6 hours; NF-κB in model lung increased as time extending in first 6 hours.3. Tongfu Decoction and Dexamethasone could decrease W/D in lung tissue, down regulate TNF-a and IL-6 content in ALI rats serum, lung and intestine; and down regulate TGF-β1 content in ALI rats lung and intestine. Also Tongfu Decoction and Dexamethasone could decrease NF-κB p65 and NF-κB mRNA expression in ALI rats lung. Results as follow:W/D, TNF-αand IL-6 content in the model group serum, lung and intestine expressed higher than the normal control group (p<0.01); and other treatment groups showed lower than the model group (p<0.01); Tongfu Decoction low-dose group showed higher than hexadecadrol group (p<0.05); both medium-dose group and high-dose group had no statistical significance(P>0.05); both medium-dose group and high-dose group were lower than low-dose group (p<0.01); medium-dose group and high-dose group had no statistical significance (p> 0.05).In lung and intestine of each group TGF-β1 contents in the model group were higher than the normal control group(p< 0.01); contents in other treatment groups lower than the model group(p<0.01); contents in Tongfu Decoction low-dose group were higher than hexadecadrol group(p<0.05); medium-dose group, high-dose group and hexadecadrol group had no statistical significance (p> 0.05); both medium-dose group and high-dose group were lower than low-dose group(p<0.05); medium-dose group and high-dose group had no statistical significance (p>0.05). Observation by light microscope showed:NF-κB p65 in the normal control group expressed negatively, while showed positive expression in the model group (p<0.01); other treatment groups showed lower than in the model group (p<0.01); NF-κB p65 in Tongfu Decoction low-dose group showed higher than hexadecadrol group (p<0.05); NF-κB p65 both in medium-dose group and high-dose group had no statistical significance(p>0.05); both medium-dose group and high-dose group were lower than low-dose group (p<0.01); medium-dose group and high-dose group had no statistical significance (p>0.05). NF-κB mRNA in the normal control group expressed thimblefully, while showed positive expression in the model group (p<0.01); other treatment groups expressed lower than in the model group (p<0.01); NF-κB mRNA in Tongfu Decoction low-dose group showed higher than hexadecadrol group (p<0.05); NF-κB mRNA both in medium-dose group and high-dose group had no statistical significance(p> 0.05); both medium-dose group and high-dose group were lower than low-dose group (p<0.01); medium-dose group and high-dose group had no statistical significance (p>0.05).Conclusions:1. Tongfu Decoction could decreased W/D, and medium-dose and high-dose group were better.2. Tongfu Decoction could decrease TNF-αand IL-6 content in ALI rats serum, lung and intestine, and medium-dose and high-dose group were better.3. Tongfu Decoction could decrease TGF-β1 content in ALI rats lung and intestine, and medium-dose and high-dose group were better.4. Tongfu Decoction could restrain NF-κB expression in ALI rats lung, which medium-dose and high-dose group showed better, and it might prevention and cure by restraining NF-κB access.
