1 ObjectiveThis experimental study uses OVA to duplicate asthma guinea pig models. We approach this subject from the effect of the Lengxiao pill playing on expiratory resistance, trachea smooth muscle, the quantity of EOS in blood and BALF, lump tissue morphology, IgE in serum, and IL-4, IL-5, NO, ET-1 in serum and BALF of guinea pigs model. And from the effect it act on allergic reaction, airway inflame-mat ion, airway remodeling and airway hyperresponsiveness, we explore its affect on treating convulsion and relieving asthma.2 Method2.1 material2.1.1 animalGuinea pig 250±50 g, male, provided by Hubei province experiment animal research center. The experiment animal licence NO:SCXK (Hubei) 2008-0005.2.1.2 Chief reagentAcetylcholine chloride; Ovalbumin; Dexamethasone; Guinea pig IgE reagent box; Guinea pig IL-4 reagent box; Guinea pig IL-5 reagent box; Guinea pig NO reagent box; Guinea pig ET-1 reagent box.2.2 Meclode2.2.1 Animals models duplicationSting 10% OVA physiological saline to the guinea pigs at one time and use 1% OVA physiological saline to fog at 15d, fogging every 30s and fogging that every 2d in 8d. Dexamethasone group, Dexamethasone lmg/kg, use the same ancount physical salt water to the original team.2.2.2.1 Test one:Observe the effect of Lengxiao pill on airway hyperresponsiveness in guinea pigs. In the proceeding of the experiment, to investigate the cavies normal situations, such as spirate, activity, diet, weight, cacation, coat, state of an illness and death, to observe the Lengxiao pill' sinfluence of allergic contract of trachea smooth muscle in vitro and expiratory resistance in guinea pigs.2.2.2.2 Test two:Investigating the Lengxiao pill influents to EOS and the lung branchus tectology of the cavy of blood and BALF suffered the asthma guinea pigs. Using the pentobarbital sodium of 3% of 30mg/kg to anaesthesia each guinea pig in each group on the 23th day, and then take the blood and wash the alveolus to collect BALF. Staining and encouting EOS in the blood and BALF, then investigating the lung branchus tectology.2.2.2.3 Test three to five:Observe the effect of Lengxiao pill to IgE of asthma cavy's serum, and IL-4, IL-5, NO, ET-1 in cavy' s of serum and BALF in guinea pigs.Take out the storing serum and BALF of cavy, reagent box of IgE, IL-4, IL-5, NO and ET-1 from refrigerator then test the index of serum and BALF according to the direction of reagent box.2.2.2 animals groupings and examination targetSixty guinea pigs are randomized into normal group, model group, Dexame- thasone group, high dosage of Lengxiao pill group, middle dosage of Lengxiao pill group, low dosage of Lengxiao pill group, every ten guinea pigs a team. To every team, the ip of medicine is 3ml, namely Lengxiao pill 16g/kg,8g/kg,4g/kg, 2.3 Statistical methods:Each group of data expresses by x±s, and uses the SPSS statistics V17.0 to process. Apply variance analysis for comparing with the mean value among groups. p<0.05 shows that the difference has remarkable statistic significance.3 ResultsGenerally, after caused allergy and arouse, guinea pig of model group appears to be decreased in activity, slow reaction, and pelage loses luster, breath times was increased, breath was difficulty. No obvious change in each pharmaceutical group.The results show that Lengxiao pill can reduce the increasing of Re and extend the trachea spiral slip in vitro (p<0.05 or p <0.01), can decrease IgE level in blood serum (p<0.01), which indicate that Lengxiao pill can remove spasm of trachea smoothmuscle and reduce asthma models'airway hyperresponsiveness definitely. It can reduce EOS counting in blood plasma and BALF (p <0.01), can decrease NO, IL-4, IL-5 level in blood serum and BALF (p<0.05 or p<0.01), suggesting that it can obviously restraint airway inflammation and inflammatory infiltration.It can decrease IgE, IL-4, IL-5 level in blood serum or BALF (p<0.01), blockage the generator segment of allergic reaction, can remarkably restraint I allergic reaction. It can decrease ET-1 level in blood serum and BALF (p<0.01), can relieve airway remodeling significantly.4 Conclusion4.1 Sensitizing and attacking by Ovalbumin can duplicate asthma guinea pig models.4.2 The attack on asthma is closely related with wind, cold, phlegm and fluid. Phlegm and fluid retention, airway spasm are the main pathogenesis of asthma.4.3 Lengxiao pill has the function of warming lung, reducing phlegm, dispersing fluid and calm panity, which can treat asthma exactly.4.4 The active mechanism of treating asthma4.4.1 Lengxiao pill can remarkably restraint I allergic reaction. The mechanism is relationshiped with its controlling IgE, IL-4, IL-5, and repress producing allergic medium.4.4.2 Lengxiao pill can obviously restraint airway inflammation. Its influence on air-flue phlogosis and decrease NO, IL-4, IL-5 level in blood serum and BALF.4.4.3 Lengxiao pill can relieve airway remodeling significantly, because it can control airway inflammation, inhibite the proliferation of smooth muscle cell, and decrease ET-1 level in blood serum and BALF.4.4.4 Lengxiao pill can reduce airway hyperresponsiveness definitely, because of decreasing IgE level, removing spasm of trachea smoothmuscle, controlling airway inflammation, and relieving airway remodeling.
