The mechanism underlining the injury of neorons casued by brain ischmia and hypoxia has been investigated deeply and widely. And many studies focused on the change on postsynaptic level.Some studies attach importance to the inducements of the injury of neurons and especially focus on the increasement of presynaptic glutamate release. However, many problems are still unclear in this field because of the complexity underlining the mechanism of the increase of presynaptic glutamate release. In this paper, we studied the relationship between the changes of presynaptic free radicals and the increase of glutamate release under chemical hypoxia with patch clamp recording method combined with biochemical and pharmacological approach. And unveil signal transduction pathway between different subtypes of presynaptic free radicals as well as that of downstream receptors or proteins, thereby figuring out the signal transduction pathway of glutamate release caused by free radicals under chemical hypoxia. The mechanism of the increase of glutamate release caused by chemical hypoxia provided clues to suggest the new presynaptic signal transduction pathway under hypoxia and theory foundations for presynaptic approach in treating stroke. Proceeding from the mechanism regarding the disorder of presynaptic signal transduction pathway, we hope to find new drugs in stroke treatment.
