mTORC2 is an important ingredient of PTEN/PI3K/AKT/mTOR signal pathway especially the excessive activation of which is closely associated with the germination and growth of the cancer so that it has become an important target of the remedy of cancer. Recently with the advent of micromolecular inhibitor clinically, mTORC2 has aroused wide interest and has been reported a lot. As a protein molecule of the mTORC2 scaffolding,Rictor maintain the stabilization of mTORC2 with other five Protein molecules. mTORC2 plays an important role in many physiological processes such as the cell survival metabolize proliferation and the compose of the cytoskeleton.But compared with mTORCl,a lot of unknown parts of mTORC2 are in need of exploration of us such as the upstream signal pathway of mTORC2, the activation of mTORC2, whether the Rictor can maintain the stabilization of the mTORC2 and the role in the mTORC2,which is an interest topic and recently there is little about the research of the unknown parts of mTORC2.If we want to study the function of the gene Rictor through the use of the biological model of zebrafish,we should prove that the zebrafish gene is conservative in evaluation and the conservative gene in the vertebrate so as to provide theoretical support for the research of the role of zebra fish in the hematopoietic system and the hemotopoietic microenvironment.The situ hybridization in the whole embryo has been widely used in the gene expression profile and the important means of explore in the function of related genes.On the basis of the rictor mRNA of zebra fish,we can compose the Antisense RNA probe and could analyze the expression of the rictor in the early stage of the development of zebra fish.And we know that the expression of Zrictor in the embryonic one cell stage and the special expression in the notochord and after 48 to72 hours special expression in the neural tube and sarcomere.Through the inhibition of Antisense morpholino oligonucleotides generation MO to Zrictor,we could observe hydropericardium the flexing of the embryo of zebra fish and the delayed development of zebra fish in the MO injection group.There is little change of the different spatio-temporal expression of zebra fish before and after the microinjection in the hematological system maker and cardiac system maker.But some mild change of Fli-1 in the intersegmental blood vessel and discounted expression of muscle and zebra fish embryo with hydropericardium after the test on formative the expression and distribute change of hemoglobin in the circle with o-staining which indicates that show reductive deposit of hemoglobin.Whether the Rictor is a fit target objective in case that the hydrone inhibitor is effective to cancer and donnot affect normal tissue ?The rictor may affect the hydropericardium and normal tissue after the inhibitation of MO in vivo so that it may become a new therapeutic target of the treatment of clinical hematopathy and cancer.
