Mitochondrial respiration imbalance was closely related with some diseases. Acutemyocardial ischemia at the top of heart troubles in appears was a typicallyrepresentative. It is foundation of drug screening for some diseases thatnatural-occurring substance or artificial synthesis regulated mitochondrial respirationso that returning to normalcy. Changes of proton leakage, respiratory ratio and oxygenutilization ratio in mitochondria were happened, KATPchannels were closed, vascularsmooth muscle was contracted, cardiac muscle were ischemia or not blood flow whenacute myocardial ischemia was occurred. Regulation of mitochondrial respirationcould be an effctive method against acute myocardial ischemia. Regulator ofrespiratory balance in mitochondria was candidating drug for acute myocardialischemia.Regulation of sulfides derived from garlic on function of mitochondrial respirationwhich related to pH, kinds of products, concentration and time was the foundation forcandidating drug against acute myocardial ischemia according as effects of sulfidesderived from garlic on acute myocardial ischemia. Since new millennium, there weresome new achievements in scientific research concerning H2S used for acutemyocardial ischemia via relaxation of vascular smooth muscle. It was intrestingresearch subject for investigators. However, taking into account toxicity of H2S andpsychological factors, directly using H2S was impossible so far. Sally port was H2Scomes from where or what for safe, stable and controllable. Garlic contained bothlipid and water solubility sulfides. Whether or not producing H2S coming garlicderived sulfides in mitochondria have no idea only on the basis of some of lipidsolubility sulfides converted into H2S via exchange of sulfhydryl group (-SH) inhuman erythrocyte and cysteine in deoxygenated hemoglobin. Therefore, these weremain researching contents that analyizing the decomposing conditions of garlicextract solution for garlic derived sulfides in different pH and extract time andelemental sulfur in appear; observing regulation of garlic extract solution, garlic derived sulfides, elemental sulfur and H2S on mitochondrial respiration balance;probing the process of garlic derived sulfides and elemental sulfur converted into H2Svia mitochondria; establishing mechanisms of garlic derived sulfides converted intoH2S via elemental sulfur and mitochondria.Objective: Changes of garlic-derived sulfides in garlic extract solution with timeand pH and conditions and times of producing elemental sulfur were explored. Effectsof garlic extract solution, garlic-derived organic polysulfides, elemental sulfur andH2S on mitochondrial respiration were observed. Feasibility and fundamental processof producing H2S via mitochondria and elemental sulfur were researched. It waspossible for H2S coming from garlic to safe, stablize and control.Results: Contents of diallyl trisulfide (DATS), diallyl disulfide (DADS),diallylsulfide (DAS) in70%ethanol extracting solution were decreasing with time,content of DADS was higher because of instability of DATS. Both allyl methylsulfide (AMS) and phenyl disulfide (DPDS) in extracting solution were not detected.Changes from DATS to DAS in alkalenscence were in favor. Elemental sulfur inextracting solution was diagnosed in order to produce H2S for regulating themitochondrial respiration balance via matrix of pH8.0with glucose in mitochondria.Under oxygening and hypoxia, rotenone as complex I inhibitor and sodiumsuccinate as substrate in FADH2short chain, effects of garlic extract solution onmitochondrial respiration balance were promoted in low concentrations and inhibitedin high. In experimental concentrations, effects of garlic-derived sulfides, such asDATS, DADS and DAS, on mitochondrial respiration balance were only promoted.Concerning promotion effects, garlic extract solution was lower than one of DADSand higher than one of DATS and DAS. Effects of garlic extract solution, elementalsulfur and H2S on mitochondrial respiration balance were similar in promoting in lowconcentrations and inhibiting in high. The differences located in concentrations forchanging from promoting to inhibiting. The decreasing orders were garlic extractsolution, elemental sulfur and H2S. State4were increased resulting leakage in protonand uncouple; decreased state3, delayed TS3-4, and depressed RCR. Effects inoxygen condition were better those in hypoxic condition in most cases.The absorption peak in670nm for producing H2S was detected for elemental sulfurwith glucose via mitochondria. These results displayed that elemental sulfur as electron donor in complex II from garlic-derived sulfides, involved electron transportin CoQ, converted elemental sulfur into H2S in mitochondrial matrix side. In sameconditions, H2S was not detected for DATS, DADS and DAS. The process ofgarlic-derived sulfides converted into H2S was complex and involved in content andactive of GSH via water solubility garlic-derived sulfides for producing H2S.Results displayed in short time, lipid solubility garlic-derived sulfides convertedinto H2S via elemental sulfur as electron donor in matrix of motochondria, involvingelectron transport in CoQ in complex II, and then inhibiting CcO. Thesegarlic-derived sulfides may be precursor of H2S for acute myocardial ischemiacandidates in the future. H2S was produced via mitochondria and elemental sulfurbesides exchange of-SH in erythrocyte and cysteine in deoxygenated hemoglobin.H2S was produced with safe, stable and controllable manners against acutemyocardial ischemia on account of different influences of garlic-derived sulfides andits catabolites on mitochondrial respiration function according to decomposingcharacteristics.Conclusions: Contents of lipid solubility garlic-derived sulfides such as DATS,DADS and DAS in70%ethanol extracting solution were decreasing with time.Changes from DATS to DAS in alkalenscence were in favor. The decreasing orders forregulating effects were garlic extract solution, elemental sulfur and H2S. Effects inaboved in oxygen condition was better those in hypoxic condition in most cases. It was safe, stable and controllable thatH2S coming garlic derived sulfides as precursor for the sake of avoiding directtoxicity of H2S and psychological factor.Innovation: Results from HPLC in garlic extract solution displayed that theprocess of DATS converted DADS in pH8.0was quicken in favor. Elemental sulfurwas detected with colorimetry method in garlic extracting solution. Effects of DATS,DADS and DAS in garlic-derived sulfides on mitochondrial respiration werefacilitated. There were dose-dependent for effects of garlic extract solution,elemental sulfur and H2S on mitochondrial respiration, facilitated in lowconcentrations and inhibited in high. It was established that the basic conditions andtime of producing H2S coming from garlic-derived sulfides via elemental sulfur inmitochondria were glucose and3-4hours. The approach of H2S cominggarlic-derived sulfides as precursor proved to be safe, stable and controllable.
