backgroundWith the rapid development of social economy, the society is in fierce competition and the pace of life increases extremely, resulting in the prevalence rate of sub-health and sub-health fatigue is rising obviously. There is a serious effect on life quality, thus, sub-health fatigue is currently a hot topic in research field.ObjectiveTo research and explore the characteristics of sub-health fatigue distribution in the Guangdong area, the risk factors of sub-health fatigue, the related quality of life and the characteristics of constitution in sub-health fatigue.To explore the biomarkers of sub-health fatigue through the urine metabonomics analysis.To analyze the rule of variation among the different constitution of sub-health fatigue.Methods 1813 cases were surveyed randomly by the sub-health questionnaire in 2010 year. Sub-health questionnaire is divided into five parts:(1) general situation:name, age, gender, nationality, marital, education, occupation and income, et al.(2) the lifestyle and experience events:including eating habits, sleep situation, exercise situation, major life event,et al. (3) Self-evaluation the sub-health state, including physical, psychological and social discomfort aspects.(4)health related quality of life (SF-36), (5)The scale of Chinese constitutions evaluationApplied'H-NMR technique with BRUKER Avance 500 MHzI NMR spectrometer with super-conducting magnet to compare the metabonomical characteristics of urine in sub-health fatigue and different constitution of sub-health fatigue and to seek biomarkers.Results1. Epidemiological study of sub-health fatigue state:①1813 questionnaires were sent,1686 were recovered,this recovery rate is 93.00%. including 1406 valid questionnaires with a qualified rate of 83.39%, according with epidemiological studies conditions. According to ask a medical history, physical examination medical examination reports and Chinese Medical Clinical Guide for sub-health,210 persons(14.94%) of disease states, remaining 1196 questionnaire(85.06%) were health and the sub-health,1196 persons were involved in the result analysis.②sub-health happens as high as 84.78% prevalently.The total prevalence rate of sub-health fatigue among was 24.4%.③Females carry the sub-health state prevalence rate is higher than men (χ2=5.625, P<0.05). There was no statistically differences in ages,educational levels,and marriages(P>0.05)④the typical manifestations of sub-health fatigue mainly involved in feeling stressed, feel live tired, feel helpless, hypomnesia,lower energy, limb fatigue, muscle aches, eyestrain, anxious, upset, depressed, poor sleep.⑤The key factors of sub-health fatigue are movement, sleep,environmental pollution,medical history,experience event.2.health related quality of life(SF-36)of sub-health fatigue①The scores of every factors in SF-36was lower in sub-health fatigue(P<0.05).②PF,BP,GH,VT and MH is associated with susceptibility of sub-health fatigue in GuangDong area.3.the distribution characteristics of constitution in sub-health fatigue:①the result showed that sub-health and sub-health fatigue were easier to tend of biased constitution than health.②In order of decreasing percentage,the rang was qi deficiency constitution (27.71%), dampness-heat constitution (24.85%), yang deficiency constitution (19.82%), phlegm-dampness constitution (19.23%), yin deficiency constitution (15.29%), qi depression constitution (17.65%), blood constitution (14.30%), the specific endowment constitution (11.24%) in sub-health③In order of decreasing percentage,the rang was qi deficiency constitution (46.56%), dampness-heat constitution (36.44%), qi depression constitution (31.98%), phlegm-dampness constitution (30.77%),yang deficiency constitution (27.94%),, yin deficiency constitution (26.32%), blood constitution (24.70%), the specific endowment constitution (15.38%) in sub-health fatigue.4.①there was obvious differences in metabolism spectrogram between control and sub-health fatigue.two groups can be distinguished obviously in sample score plot after data processing and pattern recognition.②10 metabolites' level were statistically higher than control group which might be the potential biomarkers of sub-health fatigue.③the potential biomarkers of sub-health fatigue were a-oxoglutaric acid (63.00). alanine (63.77),Coenzyme Q, (64.01),Creatinine, (63.04). Phosphocreatine (63.93),Methylhistidine (63.29).3-Phosphoglyceric acid (63.85/3.65),Guanosine (63.81),hippurate (63.97) 5.①there was obvious differences in metabolism spectrogram between control and difference constitution of sub-health fatigue, respectively. Two groups can be distinguished obviously in the sample score plot after data processing and pattern recognition.②1 metabolites' level,10 metabolites' level and 12 metabolites' level were statistically higher than control group which might be the potential biomarkers of difference constitution sub-health fatigue, respectively.③the potential biomarkers of control constitution sub-health fatigue were creatinine(63.04); the potential biomarkers of qi deficiency constitution sub-health fatigue were creatinine(63.04),Methylhistidine (63.29),glucose (63.61,3.69),alanine (63.77),Guanosine (63.81),3-Phosphoglyceric acid (63.85),Phosphocreatine (63.93),hippurate (63.97),histidine (63.25),Glyceric acid (64.13); the potential biomarkers of dampness-heat constitution sub-health fatigue were Creatinine (63.04),3-Phosphoglyceric acid (63.65,3.85),alanine (63.77),Guanosine (63.81),Phosphocreatine, (63.93),oxoglutaric acid (63.00),Coenzyme Q (64.01),glucose (63.61,3.69),Lactic acid (64.09),Aconitic acid (3.73)6.①there was obvious differences in metabolism spectrogram between control constitution and qi deficiency constitution of sub-health fatigue.there was obvious differences in metabolism spectrogram between control constitution and dampness-heat constitution of sub-health fatigue. All can be distinguished obviously in sample score plot after data processing and pattern recognition.but,there was no differences in metabolism spectrogram between qi deficiency constitution and dampness-heat constitution of sub-health fatigue.②3 metabolites' level and 7 metabolites' level were statistically higher than control constitution which might be the potential biomarkers, respectively.③the potential biomarkers were 3-Phosphoglyceric acid(63.85),hippurate (63.97),alanine (63.77) in control constitution sub-health fatigue and qi deficiency constitution sub-health fatigue.the potential biomarkers were creatinine (63.04),oxoglutaric acid (63.00),Coenzyme Q(64.01),3-Phosphoglyceric acid(63.65),alanine (63.77),adenosine (64.81),vanillylmandelic acid (64.85) in control constitution sub-health fatigue and dampness-heat constitution sub-health fatigue.conclusion1.The prevalence rate of sub-health fatigue, the correlated factors are very complicated, survival quality is obviously decreased.sub-health fatigue was more reaction by SF-36 scale and sub-health questionnaire.it is necessary to carry out different effective intervention to prevent and treat sub-health fatigue in many ways,in order to improve survival quality.2. sub-health and sub-health fatigue were easier to have tendency of biased costitution than healthy persons.In order of decreasing percentage,the rang was qi deficiency constitution, dampness-heat constitution, yang deficiency constitution, phlegm-dampness constitution, yin deficiency constitution, qi depression constitution, blood constitution, the specific endowment constitution in sub-health.In order of decreasing percentage,the rang was qi deficiency constitution, dampness-heat constitution, qi depression constitution, phlegm-dampness constitution,yang deficiency constitution, yin deficiency constitution, blood constitution,the specific endowment constitution in sub-health fatigue.3.Sub-health fatigue is highly correlated with energy metabolism confusion.Biomarkers can be used to diagnose and evaluate sub-health fatigue.4.There are common biomarkers and difference biomarkers among difference constitution of sub-health fatigue.5. there was obvious differences in metabolism spectrogram between control constitution and biased constitution of sub-health fatigue. Form the metabonomics point view, we explored the biological characteristics of the different constitutions, which might contribute some new explanation for manifestation characteristics, forming factors and mechanism of the control and biased constitutions.
