Objectives:To investigate the change of metastasis ability of A549cell through the RNA interference of RACK1; And to briefly investigate the mechanisms of RACK1facilitating the metastasis of lung adenocarcinoma; Then study the expression level of RACK1between prognosis and clinico-pathological factors of lung adenocarcinoma.Methods:Human lung adenocarcinoma cell A549were transfected with4RACK1shRNA plasmids. Real-time PCR, RT-PCR and Western-blot was introduced to investigate the interference efficiency. Transwell migration and invasion assay, wound healing assay and immuno-fluorscence of cytoskeleton was introduced to study the function of RACK1in facilitating the abilities of A549cell in metastasis and invasion. The downstream molecules of RACK1, including Snail, E-Cadherin, N-Cadherin, Vimentin and MMP-9, was examined by Western-blot. Finally, the expression of RACK1in human lung cancer clinical samples was examined by Immunohistochemistry; further relationship of RACK1expression and the prognosis and clinico-pathological factors of lung adenocarcinoma was investigated.Results:shRNA1of4RACK1shRNA plasmids was found to show greatest interference efficiency. Both transwell migration and invasion assay, wound healing assay provided with same result that the interfering of RACK1, A549RACK1-shRNA1, inhibited the abilities of metastasis and invasion of A549cell. Immunoflourscence of cvtoskeleton showed that the surface of A549RACK1-shRNA1cell is irregular and the micro skeleton of the cell is massed up. The expression of Snail, N-Cadherin, Vimentin and MMP-9was significantly decreased in A549RACK1-shRNA1cell, meanwhile the expression of E-Cadherin was significantly elevated in A549RACK1-shRNA1cell. Finally in the over-all survival analysis and disease-free survival analysis, the median survival time of RACK1high expression group was significantly lower than the RACK1low expression group. The RACK1expression was significantly correlated with TNM staging and lymph node metastasis. RACK1high expression is a independent risk factor of the poor prognosis of lung adenocarcinoma.Conclusions:The study investigated that RACK1could promote metastasis of lung adenocarcinoma in vitro, and briefly examined the downstream molecules of RACK1. We also found that RACK1high expression in human lung adenocarcinoma is correlated to poor prognosis and correlated to many clinicopathological factors. RACK1high expression in human lung adenocarcinoma is an independent risk factor of the poor prognosis of lung adenocarcinoma.
