Hair follicles are developmentally distinguished as hair germs formed by a series of interactions between the embryonic ectoderm and the mesoderm. These interactions continue during the development, growth, and differentiation of hair follicles. Although not experimentally proven, progenies of the interacting mesodermal cells are believed to be incorporated into a specialized portion of the follicle (hair bulb) as dermal papilla cells. Dermal papilla locates in the bottom of follicle. Hair matrix cells are around it. It is mainly composed of dermal papilla cells(DPC).Using transplantation technique demonstrated that DPC is specialized mesenchymal cells which can induce hair follicle formation during hair follicle development stage and take a center role in hair follicle growth and regulation of growth cycle. It is difficult to isolate and culture dermal papilla cells because dermal papilla is in a special position. In order to find the simple method of isolation and cultivation of dermal papilla cells, dermal papilla were digested from tissue with two steps enzyme method and purified by centrifugation. Isolating time was shortened and working strence was decreased, the harvest rate of DP was increased with this method. Growth pattern, expression of a -actin, and the effect of non-serum culture of dermal papilla cells were observed. Dermal papilla is mainly composed of dermal papilla cells(DPC) which expressed a -actin. It has many functions in regulation hair growth and inducement hair formation. The contains and structure of DP are changed in hair growth cycle. Hair matrix cells in bulb can activate DP. It抯 structure and volume HI had been changed. It demonstrates that a serious of interactions between mesenchymal and epithelial had been taken place. Growth factors, adhere molecules, polysaccharides increased or decreased during hair follicle cycle. But their functions are unclear. To demonstrate the interactions of dermal and epidermis, to understand the control of hair follicle growth cycle and to find out a pathway to reconstitute hair follicle in vitro and in vivo, human dermal papilla cells and human hair follicle epithelial keratinocytes co-coltured in DMEM with 10% FBS or no-serum. The cells were counted in different time and the cells growth pattern was observed as well. Hair follicle is the complex affiliated organ of skin. More than 20 kinds of cell populations, which come from dermal and epidermis take part in the course of hair follicle formation. The complex interactions between those cells and extra cellular matrix influence the functions and signal transduction of the cells that is interfered by cellular surface molecular. DPC is a center cell to control hair follicle growth. It can regulate the differentiation of hair follicle, The results of hair follicle cell graft research demonstrated that only DPC can induce hair bed or epithelial to form hair follicle structure in vitro and in vivo. The interactions between the cells are very complex in vivo. Many factors may be involve in the course. To investigate the regulation activity of mesenehymal cells to differentiation of epithelial cells from hair follicle and to study its differentiation property, mesenchymal cells gel was made by nubby dermal papilla cells, free dermal papilla cells, skin fibroblasts.
