Molecular CD 147, namely EMMPRIN (Excellular Matrix Metalloproteinase Inducer).It can stimulate fibroblasts to produce Matrix Metalloproteinase (MMPs). MMPs is closely related to invasion of tumor because it can digest the excellular matrix. We want to construct antisense RNA expression vector of CD 147 to inhibit the expression of CD 147 and secretion of MMPs, then to explore its function of inhibiting invasion of tumor.Four parts are included in the experiment process. 1. Identifying expression of CD 147 on ovarian cancer cells.2.Constructing the antisense RNA expression vector of CD 147 .S.Transfecting ovarian cancer stain 8910. 4. Observing its function of inhibiting invasion of ovarian cancer cells. After identifying expression of CD147 on ovarian cancer cells.,we cut the pbluescript sk(+)CD147(containing full length of CD 147 cDNA ) with two enzymes and obtain 1.7kb CD 147 cDNA,then. link the cDNA reversely to mammalian expression vector PCDNA3.1 (+) cut by two similiar enzymes too.After that,\ve transfect the antisense RNA expression vector to ovarian cancer strain 8910 which express CD 147 highly.After selecting by G418, picking out single clone by method of limited dillution ,enlargly cultuing,we identify the clones by immuno-histochemical SP method, cytoflowing method , RNA dot blotting and observe states of inhibiting proteinexpression.Gelatinase Zymography is used to study the function of CD 147 in aspect of inducing secretion of type IV collagenase;cell chamber is used to test the function of inhibiting invasion of ovarian cancer cells.The result shows the vector is constructed successfully;expression of CD 147 is blocked efficiently;secretion of type I\ collagenase is reduced;invasion of ovarian cancer cells are lowered.Conclusion: we inhibit the expression of CD 147 through molecullar level ;and .because of this.its capability of stimulating human fibroblasts to secret MMPs is lowered;at last.it indirectly inhibit the infiltration and metastasis of ovarian cancer cells.
