OBJECTIVE: To investigate the mechanism of the Bujingheji (BJHJ, a kind of patent traditional Chinese medicine) which has the effect of regulating menstruation to treat the delayed menstruation or accompanying pomenorrhea. METHODS: Adopting the female presenile rats as the natural kidney-deficient animal models, and establishing 5 groups as follow: the high and the low dosage groups of the BJHJ, the WJBFW (a kind of patent medicine has a similar effect) group, the blank test groups of the presemle and young rats. After lasting 15-day intragastric (i.g.) medication with the BJHJ, the author applies the fluorospectrophotometry, the radio-immunity analysis and the immunohistochemistry to observe the changes happened in the hypothalamus-pituitary-ovary(H-P-0) axis of the animal models, such as the monoamines neurotransmitters in hypothalamus the sex hormones in blood serum, the pathomorphology of uterus and ovary, and the estrogen receptor(ER) and progestogen receptor(PR) and follicle-stimulating hormone. receptor(FSHR), and the apoptosis markers as Fas, caspase-3 and Ki67 in uterus andovary in order to research the menstruation of the effects of the BJHJ in cellular or molecular, levels.RESULTS: The BJHJ can obviously decrease the contents of norepinephrine (NE) N dopamine(DA) in hypothalami of the presenile rats; And it can increase the level of estradiol(E2) and progestogen(P) in blood serum; Furthermore it can increase the numbers of ovarian follicle and corpus luteum, the depths of mucous membrane and the bore of gland; In the same time, it can rise the expression of FSHR on follicle and corpus luteum and ER, PR on uterus; Also it can restrain the apoptosis expression of caspase-3 on follicle cells; On the contrary, it has no clear effects to these markers, such as: Fas, Ki67 and insulin-like growth factor-I( IGF-I).CONCLUSION: Summarizing the above-mentioned results, we can get the conclusion that the BJHJ has the effects to regulate the female H-P-O axis by acting on multiple pathways and effectting multiple targets, maybe this is the mechanism of the BJHJ to treat the delayed menstruation or hypomenorrhea.
