Preliminary Analysis of Gene Expression Profiles in Human Myocardium of Atrial Septal Defect by cDNA MicroarrayObjective To investigate the difference in gene expression profiles between auricular myocardium of atrial septal defect (ASD) and normal auricular myocardium using cDNA microarray, and to analyze the feature of gene expression profiles in human ASD myocardium. Methods The cDNA probes prepared from mRNA of auricular myocardium of ASD and normal auricular myocardium was hybridized with two identical cDNA microarray membranes containing 18000 human genes. The functions of the differentially expressed genes were analyzed by bioinformatics. Results (1) 1056, 264 and 47 genes were found significantly differentially expressed over 2, 3 and 5 folds respectively between auricular myocardium of ASD and normal auricular myocardium. (2) There are 130 ESTs ( Expressed Sequence Tag ), 248 function-unknown proteins or hypothetical proteins, 678 Unigenes with known function in the differentially expressed genes. (3) Different clusters of genes associated with embryo development, cell differentiation, metabolism, signal transduction, DNA transcription, cell skeleton, ion channel, apoptosis, protein metabolism, immune, etc were concerned. Conclusion Impaired signal transduction including Wnt signal pathway, expressive upregulation of extracellular matrix-related genes, dysregulation of cell cycle, initiation of fetal transcription program, higher requirement of energy, derangement of cell skeleton, reduced contraction capacity of myocardium, caused apoptosis of auricular myocardium, increased synthesis of proteins with cell protection, decreased anti-virus immunity, expressive abnormality of Nkx2.5-downstreamed genes, and gathered chromatosome map (17pl3,19pl3.1-13.3, 19ql3.1-13.4,21q22.2-21q22.3,6p21.3) of many differentially expressed genes, etc are the features of gene expression profiles of auricular myocardium of human ASD.
