The Role Of Of Ephedra The Forsythia Chixiaodou Soup Anti-type I Allergy And The Mechanism Of Experimental Research

Objective:The major phases of type I allergy disease include sensitization and triggering.The high concentration of the special IgE has important effect in the sensitization ,and the degranula of mast cell in the triggering is important too. According to the pathogenesis of type I allergy disease, The study utilize serologic pharmacological method, watch the vary of the production of special IgE and the protein activity and gene expression of Tul/T^ cytokine and apoptosis of T lymphocyte and degranual of mast cell and the release of histamine. In order to investigate the mechanism systemically of mahuanglianqiaochixiaodou soup in the treatment of type I allergy disease.Methods: l.The effect of mahuanglianqiaochixiaodou soup on the special of IgE: use type 721 spectrophotometer to watch the OD610 values of absorption of the Ceruleus in the Passive Cutaneous anaphylaxis(PCA) .2.The effect of mahuanglianqiaochixiaodou soup on T lymphocyte and related Tul/T^ cytokine : First make blood serum which contain mahuanglianqiaochixiaodou soup, then use gas chromatograph (GC) to qualitative analysis if the serum contains some Chinese herbal medicine. The study was divided into six consist of groups: normal group, model group, 5%serum-treated group, 10% serum-treated group and western medicine-treated group. The mice in each group were sensitized with OVA exclude in the normal group, Then took out the spleen of each mouse in germfree condition, separated T lymphocyte, added ConA, OVA and related sera partly. After cultured 72h, collected T lymphocyte cells and supernatant separately. The protein content of IL-4, IL-13 and IFN- Y in the supernatant were detected by Enzyme-linked immunoadsordent assay (ELISA) ,The mRNA of IL-4, IL-13 in the T lymphocyte cells were detected by in situ hybridization method , and the apoptosis of T cell were detected by electron microscope and TUNEL. 3. The effect of mahuanglianqiaochixiaodou soup on the degranulation of mast cell: watched the number of degranulated mast cell, and the content of histamine were detected by FluorescenceResults: 1. Inhibit the production of the special IgE: the results of the PCA experiment showed Ceruleus were dwindled significantly (P<0.01or 0.05), There was no remarkable difference in OD610 values of absorption between 10% serum-treated group and the western medicine-treated group(p>0.05). the result imply mahuanglianqiaochixiaodou soup can inhibit type I allergy reaction probablythrough inhibit the production of special IgE. 2.Provide and detect the serum which contain drug: The qualitative analysis of GC in the serum shows the traditional Chinese medicine sera had efedrina,but the normal sera hadn't. So The provided sera can be used to do the next experiment. 3.The effect of protein activity and mRNA gene expression of TH1fTH2 cytokine :The result of ELISA showed mahuanglianqiaochixiaodou soup can decrease the content of IL-4(P<0.01or 0.05) and increased the content of INF- Y (P<0.01or 0.05), but have no significal effect on the content of IL-13 (P> 0.05). The result of in situ hybridization showed mahuanglianqiaochixiaodou soup can decrease the content of IL-4mRNA (P<0.01or 0.05) and increased the content of IFN- Y mRNA (P<0.01or 0.05). There was no remarkable difference between 10% serum-treated group and the western medicine-treated group(p>0.05).The function that mahuanglianqiaochixiaodou soup can inhibit the synthesis of IgE probably correlated with the inhibitation of TH2 type cytokine and acceleration of TH1 cytokine 4.The eftect of T lymphocyte :The result of electron microscope and TUNEL showed T lymphocyte occurred transparent apoptosis .Binding above-mentioned experiment ,We can presume this apoptosis cell may be TH2 type T lymphocyte cell. S.The effect on mast cell degranulating and histamine releasing: mahuanglianqiaochixiaodou soup can inhibit mast cell degranulating and reduce the relation of histamine.Conclusion: mahuanglianqiaochixiaodou soup can inhibit the production of the specifical IgE in the serum, and inhibit the synthesis...
