Objective: Atherosclerosis (AS) can occurred in Aorta, Coronary, Cerebral arteries, Renal arteries, Mesenteric arteries and Arteries of extremity. Coronary sclerosis and cerebral arteriosclerosis are the most common diseases and most dangerous to human health. Recently researches on the injure mechanism of AS especial about the role of endothelial cell injury in the occurrence of AS have been widely done, but effective drugs of protecting endothelial cells, preventing AS and reducing sclerosis plaque have rarely been reported. Chinese Medicine consider that the pathogenesis of AS must have relations among cardio-gas-weak, blood stagnation and sputum. Xiong-bi-tong fang(XBTF) composes of mending Qi, activating blood and dissolving sputum. It has significant effect on hyperlipemia and cardiac angina in clinic, but there isn't experiment data to explain its mechanism. The paper mainly try to find out the anti-AS mechanism of XBTF from protecting endothelial cells and blocking initiation links of AS. These will provide theoretical and experiment support for the usage of the prescription on AS.Methods: Sixty rabbits were randomly divided into six groups, AS control group( I ), Fu fang dan shen piang (FFDSP) group( II), Xiongbitong small dose group(III), Xiongbitong middle dose group(IV), Xiongbitong big dose group( V) and normal group(VI). Group VI were fed with normal feedstuff and other groups were fed with high-cholesterol feedstuff to cause AS. Group II III IV V were treated with drugs. At the end of experiment the pathologic changes were checked under light microscope and electronic microscope. Many bio-markers such as TC, TG, HDL-C, LDL-C, apoAl, apoB, NO in serum and blood vessel walls, ET-1 in plasma and blood vessel walls, plasma t-PA and PAT, NOS classification and activity, exfoliated endothelial cell number, apoptotic rate of endothelial cells, Bax, Bcl-2 were measured.Results:1) The AS pathologic changes can be observed on every rabbit at AS control group, commonly occurred at branch of aorta and descending aorta, the pathologic changes were pin-like blotches and unfixed length yellow striations on the endothelial surface, some gray-yellow plaques that apparently projected on the endothelial surface also can be observed. Under the light microscope the hyaline degeneration of endothelial connective tissue, cap-like projection, vacuolation of endothelial smooth muscle cells, a few fragment of necrosis cells and WBC can be observed Under the electronic microscope the un-integrity blood endothelium, apparently proliferated blood endometrium, deposited fibro-protein, mass proliferated smooth muscle cells can be observed. The pathologic changes at Xiongbitong three-dose groups were ameliorated at different levels.2) High-cholesterol feedstuff can cause rabbits' hyperlipemia, at AS control group the value of TC, TG, LDL-C and apoB were raised, odds of HDL-C/LDL-C and apoAl apoB were apparently reduced (P<0.01). The values of TC, TG, LDL-Cand apoB at Xiongbitong three-dose groups were reduced at different levels and odds of HDL-C/LDL-C and apoAl/apoB were adjusted. There is a marked dose-effect relationship. The effect of FFDSP was similar with small dose group.3) Results showed that at AS early stage the Values of NO, tNOS and ENOS were apparently reduced but the values of ET-1, IONS and odds of INOS/ENOS were remarkably raised, p values were less than 0.01 compared with normal group. The values of NO. tNOS, ENOS at Xiongbitong three-dose groups were raised at different levels, the values of ET-1, IONS were reduced and odds of INOS/ENOS were adjusted. There is a marked dose-effect relationship at those biomarkers. The protect effect of FFDSP on eNOS was less than small dose group.4) The value of plasma t-PA at AS control group was apparently reduced but the value of PAT was remarkably raised, so the odds of t-PA/PAT markedly reduced.
