Objective: To research the heredity and immunity mechanism of electro-acupuncture on rheumatoid arthritis(RA) by observing the effect of electro-acupuncture on HLA-DR4 transgenic RA animal model.Methods: The type II collagen-induced arthritis(CIA) in HLA-DR4β 1 transgenic mice, negative transgenic mice and non-transgenic mice was taken as research subjective. Every kinds of mice was random divided into electro-acupuncture group, methotrexate group and model contrast group. The feature and mechanisms of electro-acupuncture on RA were detected by using RT-PCR, Northern blot, Southern blot, dot blot, radio-immunoassay and pathologic methods.Result: firstly, electro-acupuncture could significantly attenuate inflammation and increase the pain threshold of CIA mice. Secondly, electo-acupuncture could inhibit the activities of T cell and NK cell. Thirdly, electro-acupuncture could regulate the expression of RA predisposing genes at some degree.Conclusion: The electro-acupuncture therapy treated CIA mice trough regulating the expression of RA predisposing genes and influencing the antigen presentation of MHCII moleculars.
