1 ObjectiveTo develop the experimental rat model of post operative fatigue by partial resection of the liver that simulate the human post operative fatigue (POF) and lay groundwork for the ultimate establishment of the experimental rat model and screen of herbs for POF.2 MethodAccording to the clinical symptoms and manifestations of POF, the rat model of POF was developed by the partial resection of the liver and those behavioral changes prior and post to operation, the disorder of nutritive intake after operation, stress reaction (pathological change of mucous membrane in small intestine) and the hepatic albumin gene expression would be observed. On the basis of the previous research, decoction of anti-POF was used to cure the rat model of POF.3 Result3.1 To develop the experimental rat model of post operative fatigu1. Changes of behavior: After the 70% resection of the liver, typical features appearing in the rat model was that narrowing eyes, falling in physical strength and activity, erecting hair and the loss of luster in hair, dirt attachment around annuls, eyes and noses. Signs such as low body temperature, phricasmus and ice-cold in extremities and lower sensitivity and reactivity also found.2. Changes of the indexes about nutrition: The serum levels of the iron, total protein of the liver, albumin, globulin, hemoglobin and erythrocytes and so on as the index of nutrition obviously dropped, the difference was significant in the levels compared with sham-operative group.3. The changes of the mucous membrane in small intestine: Widely congestion, edema and exfoliation of the most villi even necrosis in the mucous membrane was seen in the pathological section of POF model group while the mucous membrane in small intestine from sham-operative group was intact with light edema, the exfoliation of the villus top and not obvious congestion in lamina propria. It indicated that the injury of the mucous membrane was resulted from the stress reaction after the resection of the liver.4. Hepatic albumin gene expression: The expression of the albumin was weak in the model group treated with distilled water. The brightness of the 776 bp band was obviously weaker than the other three groups. It suggestedthat the hepatic albumin gene expression reduced after operation and that was one of causes resulting in the loss of albumin.3.2 Intervention of decoction of anti-POF in rat modelThe decoction of anti-POF had the obvious effect of intervention. It could correct, alleviate and improve the rat model in typical signs, and increase the level of the indexes about nutrition, alleviate the injury of the mucous membrane in small intestine caused by stress reaction, improve the expression of the albumin gene, all of which reduced the occurrence of POF.On the basis of the result from experiment 1-3, it concluded that the rat model of POF accorded with clinical POF in symptoms such as lassitude, drowsiness, depression and apathy and simulated well the occurrence and development of POF, it could be used for research about POF.Three factors are involved in the mechanism of anti-fatigue: 1 .protecting or repair the mucous membrane of small intestine to increase the intake of nutrition.2.improving and promoting the circulation of blood to increase the blood and oxygen supply and excretion of the metabolite.3.upregulating the gene expression of albumin to increase the content of protein.4 ConclusionThe rat model of POF according with the human post operative fatigue syndrome in symptoms can be developed by 70% resection of the liver. The decoction of anti-POF can effectively relieving the clinical symptoms of rat model of POF. It concludes that the experimental rat model of POF by partial resection of the liver can be used for the evaluation and screen of herbs for POF.
