Anxiety is one of the physiopsychic diseases with high incidence rate.This disease is due to individual characteristic and social psychicfactors in the view of modern medicine. The major treatment by modernmedicine is medicine intervention, but current anti-anxiety medicine isof shortcomings as dependable, postponed effect, interaction betweendrugs and side effect. To take the advantage of the characteristic ofChinese medicine formula, such as multi-componentscoexistence,mult-target, mult-pathway, it is impossible to find new anti-anxietyChinese medicine. But there is not so much involving anxiety animal modeland the appraisal criteria of drug effect, the mechanism and modern meaningof preventing and treating anxiety by Chinese medicine from the experimentstudy. This topic adopts the international universal anxiety animal modeland known pathological and physiological mechanism of anxiety. The basisis the experience of preventing and treating anxiety by Chinese medicineand of the way of making principle and formula. The Suanzaorendecoction(SZRT) is chosen. On the basis of confirming its anti-anxietyeffect, the best dosage is established, meanwhile, the possible endocrineand immune and condition of the animals with EPM model is explored. Thepossible mechanism of anti-anxiety effect of SZRT is probed from the animal,cell, molecule levels from the aspect of nervous-endocrine-immunenetwork. This dissertation contains the documents research and the experiment.The clinical and experimental research of SZRT, the pathogenesis ofanxiety and nervous-endocrine-immune network are summarized in thedocument part. The appraisal of dose response relation about praxiologyof anti-anxiety effect of SZRT, the exploration of endocrine and immunecondition of animal with anxiety model, regulatory mechanism ofanti-anxiety effect of SZRT in respect of nervous-endocrine-immunenetwork are discussed in the experimental part. Experiment 1:The appraisal of dose response relationship on6 酸枣仁汤抗焦虑作用的神经-内分泌-免疫网络调节机制anti-anxiety effect of SZRT in praxiology Mechods: the rats are divided into model, DZP, SZRTⅠ,Ⅱ, Ⅲ, Ⅳ sixgroups randomly, 15 rats every group. SZRTⅠ,Ⅱ, Ⅲ, Ⅳ groups areadministrated orally with 3.75g/kg/d, 7.5g/kg/d, 15g/kg/d, 30g/kg/drespectively, DZP group 1mg/kg, model group equivalent saline, they areadministrated 10 days continuously, the praxilological test, observation,and comparing various praxiological index on the tenth day. Results: SZRTis of certain anti-anxiety effect in dosage of ranging from 7.5g/kg to15g/kg, in conformity with DZP in effect direction. According to variousindex in EPM rats, the 7.5g/kg dosage is the best; 30g/kg SZRT can increaseOE+CE significantly, improve the rearing value in the sealed branch,showing certain sedation effect. Conclusion: Chinese medicine SZRT doeshave anti-anxiety effect in the dosage of ranging from 7.5g/kg to 15g/kg,we consider this effect has no relationship with sedation and hypnosiseffects, since it does not effect the motor ability; when reaching certaindosage, the effect SZRT will not improve according to the dosage, butshowing certain sedation effect. The acting characteristic of SZRT thatthere is anti-anxiety effect in lower dosage , while sedation effect inhigher dosage implicates the Chinese medicine compound might be ofdifferent clinical effects in different dosage. Experimental 2: Exploration on endocrine and immune condition ofanimal with anxiety model Methords: to observe and test the IL-2 and ACTH change in the serumof 6 group of rats at different time points(namely 0h, 3h, 6h after theEPM test, control and EPM groups). Results: the serum IL-2 level improvedsignificantly immediate after the EPM test, decreased significantly inthe subsequent 3-6h, the trend was it was lower than normal level; theplasm ACTH level had no significant change since immediate after the EPMtest to 3h after...
