1 Clinical research1.1 ObjectiveTo observe the therapeutic effect of Gu Ben Xiao Liu capsule (GBXLC) in the treatment of Advanced Non small cell lung cancer (ANSCLC).1.2 MethodsThe way of partial prospect randomized-control research was adopted. 139 Patients with ANSCLC were allocated randomly into two groups, they were treated by chemotherapy alone and chemotherapy plus GBXLC. Another 139 cases who refused to accept chemotherapy were collected as control.1.3 Results(1)The response rate of GBXLC, chemotherapy alone and GBXLC plus chemotherapy were 4.4%, 15.5% and 30.9% respectively. The response rate in GBXLC plus chemotherapy was higher than that in chemotherapy alone (P<0.01). The rate of CR+PR+NC in both of GBXLC and GBXLC plus chemotherapy was better than that in chemotherapy alone(P<0.01). (2)Main symptoms and quality of life (QOL): As to the relieving of main symptoms such ascough, fatigue, breathlessness, phlegm and dark red tongue, the therapeutic effects in both of GBXLC and GBXLC plus chemotherapy were better than that in chemotherapy alone(P<0.05, P<0.01 respectively). Compare to the chemotherapy, the dropping level of KPS in GBXLC and GBXLC plus chemotherapy was not so seriously as that in the chemotherapy group(P<0.01). (3)GBXLC could relieve neurotoxicity and gastrointestinal reaction causedby chemotherapy. As well as it could protect the function of marrow. (4)Long-term survival: Kaplan-Meier analysis showed that the median survival in both GBXLC and GBXLC plus chemotherapy were 13 months, longer than that in chemotherapy alone(9 months), P<0.05 respectively. Both 1-year and 2-year survival rates in GBXLC and GBXLC plus chemotherapy were higher than that in chemotherapy alone, p<0.05 and P<0.01 respectively.1.4 ConclusionGBXLC might inhibit the progression of tumor, promote the quality of life and prolong the long-term survival. Furthermore, GBXLC might enhance therapeutic effect and relieve the side effect of chemotherapy.2 Experimental research2.1 ObjectiveTo study the inhibitory effect and possible mechanism of herbal medicine Gu Ben Xiao Liu capsule (GBXLC) and its main functional components on A549 adenocarcinoma of human lung.2.2 Methods80 athymic mice bearing A549 adenocarcinoma of human lung were randomly divided into 8 groups: normal saline group(Control), cytoxan group (CTX), high dose group of GBXLC(H- GBXLC),low dose group of GBXLC(L- GBXLC), high dose of GBXLC plus cytoxan group(H+ CTX), low dose of GBXLC plus cytoxan group(L+ CTX), tonifying Qi components group (TQ) and promoting blood flow and detoxicating components group(PD).54 days after inoculating A549, the mice were sacrificed by dislocating cervical vertebra. The body weight, the carcinoma weight and inhibitory rate werecalculated.HE staining and TUNEL staining were performed to study histology changes and apoptosis of carcinoma tissue.2.3 ResultsThe results showed that the average weight of carcinoma in Control was 2.73±0.49g, 1.25± 0.35g in CTX, 1.7±0.36g in H- GBXLC, 1.83±0.52g in L- GBXLC, 1.16g±0.26g in H± CTX, 0.93±0.21g in L± CTX, 1.65±0.64g in TQ and 1.82±0.59g in PD. The inhibitory rate in posterior 7 groups were 54.05%, 37.66%, 32.97%, 57.63%, 65.75%, 39.42% and 33.37% respectively. Either H- GBXLC or L- GBXLC inhibited carcinoma growth obviously, compared with Control, .P<0.01 respectively. L± CTX had a higher inhibitory rate than CTX, P<0.05.Microscopic observation found that there were tissue necrosis in each group and its average area in Control was 53.42%, 49.58% of CTX, 55.44% of H- GBXLC, 30.83% of L- GBXLC, 23.33% of H± CTX, 20.83% of L± CTX, 58% of TQ and 47% of PD.TUNEL staining showed that positive expression of apoptosis in each group. Control was 32/64, H± CTX 27/30, TQ 27/30 and PD 17/25. The apoptosis index was 0.79 of H± CTX, 0.89 of TQ and 0.86 of PC respectively.2.4 ConclusionGBXLC remarkably inhibits A549 adenocarcinoma growth of human lung, as well as it has the synergism with cytoxan. It is possible that it induces carcinoma apoptosis and tissue differentiation.
