Objective: The γδT lymphocytes have close correlations with tumor immunity. In this paper, we have made a model of transformation of B-lymphocytes infected by EB virus in order to study the changes of several membrane molecules and antigenic sites after transformation .So we may find some tumor antigens recognized by γδT lymphocytes. The long goal of our research is to find some tumor antigens recognized by γδT lymphocytes ,to understand the effect of EB virus in carcinogenesis and may be benefit to diagnosis and treatment of tumor. Methods: 1. Culturing B-lymphocytes with EB viruses was to make model of transformation of B-lymphocytes by EB viruses. 2. Test expression of hsp60 and hsp70 on the surface of B-lymphocytes and EB virus transformed B-lymphocytes by cytofluorometry. 3. Using MTT method ,we detected the cytotoxicities of B-lymphocytes and EB virus transformed B-lymphocytes by γδT lymphocytes. 4. We used anti-hsp60 antibody and anti-hsp70 antibody to block cytotoxicites of γδT cells to EB virus transformed B-lymphocytes. 5. Extracting total RNA from specimens; RT-PCR was exploited to study the expression of MICA gene in B-lymphocytes and EB virus transformed B-lymphocytes. Results: 1. EB virus can transform B-lymphocytes and B-lymphocytes of PBMC. But transformation of blood by EB virus was failed. 2. Compared to normal control, almost all EB virus transformed B-lymphocytes expressed hsp60 and hsp70. 3. One's own γδT lymphocytes had a little cytotoxicites to B-lymphocytes, but more than ten folds cytotoxicites to EB virus transformed B-lymphocytes. The other's γδT lymphocytes had notable cytotoxicites to B-lymphocytes and EB virus transformed B-lymphocytes. 4. The cytotoxicity of γδT lymphocytes to EB virus transformed B-lymphocytes was markedly blocked by anti-hsp60 antibody and anti-hsp70 antibody. 5. Compared to normal control, EB virus transformed B-lymphocytes expressed MICA gene. Conclusion: Transformation of B-lymphocytes by EB virus was successful, but transformation of blood was difficult to success. Compared to normal control, EB virus transformed B-lymphocytes expressed more hsp60 and hsp70. The γδT lymphocytes had notable cytotoxicites to EB virus...
