Study on the distribution of neurochemical components in the olfactory bulb (OB) is important to study their function in the olfactory system. Lesions of olfactory epithelium (OE) can lead to the deafferentation of the OB, investigating the effect of deafferentation on the OB is helpful to discuss the mechanism of olfactory dysfunction. By immunohistochemistry, we detected the immunohistochemical localization of tyrosine hydroxylase (TH) , olfactory marker protein (OMP) , calretinin (CR) , parvalbumin (PV) , calbindin D-28K (CB) , ErbB-3 and ErbB-4 in the OB of adult Sprague Dawley rat, and OMP, CR, PV, CB, ErbB-3, ErbB-4 and c-fos in the postnatal developmental ICR mice OB. Furthermore , we studied the effect of deafferentation induced by intranasal irrigation with chemical reagent (ZnSO4 or Triton X-100) on the expression of these proteins in the OB of adult SD rat.Result:1. Immunohistochemical localization of TH, OMP, CR, PV, CB, ErbB-3 and ErbB-4 in adult SD rat OB.TH: TH-immunoreactivity (IR) were mainly present in the GL, some in the EPL.OMP: Only the ONL and GL were OMP- IR.CR: CR were localized in all layers except the ONL.PV: PV were mainly present in the EPL, few in GL.CB: CB were mainly localized in the GL, some in the EPL and IPL.ErbB-3: ErbB-3 were mainly found in the IPL, GCL and SEL, some in GL and EPL.ErbB-4: ErbB-4 were mainly found in the MCL, EPL and GL.2. Immunohistochemical localization of CB, CR, PV, ErbB-3, ErbB-4, c-fos and OMP in postnatal developmental ICR mice OB.(Postnatal day 10,20, 40: P10, P20, P40).CB: At all ages, CB-IR were predominately localized in the GL, while immunoreactivity was rarein the EPL, GCL and SEL.CR: At all ages, CR-IR were localized in all layers of the mice OB.PV: PV-IR cells were mainly present in the EPL of P20 and P40, while not in P10. Some in theONL and GL in all ages.ErbB-3: No ErbB-3-IR was found in P10 OB. MCL was weakly stained by ErbB-3 in P20 OB.AtP40 many ErbB-3-IR cells were found in the IPL, GCL and SEL, few in the EPL and GL.ErbB-4: At all ages, ErbB-4 was detected in the MCL, EPL and GL.c-fos: At all ages, c-fos was detected in the GL, MCL and GCL.OMP: At all ages, OMP was detected in the GL and ONL.3. Effects of ZnS04-lesioned olfactory epithelium(OE) on TH expression of adult rat olfactory bulb(OB).10, 20, 30d after ZnSO4 treatment, the number of TH-IR cells of experimental OB decreasedgradually, the result of 60d was similar to that of 30d. It indicated that the lesion induced by ZnSO4 was irreversible.4. Effects of ZnSCj-lesioned olfactory epithelium(OE) on CB expression of adult rat olfactory bulb(OB).10, 20, 30d after ZnSO4 treatment, the number of CB-IR cells of experimental OB decreased gradually, the result of 60d was similar to that of 30d. It indicated that CB expression in the adult rat OB was affected irreversibly by deafferentation.5. Effects of ZnSCVlesioned olfactory epithelium(OE) on PV expression of adult rat olfactory bulb(OB).10, 20, 30d after ZnSO4 treatment, the number of PV-IR cells of experimental OB decreased gradually, the result of 60d was similar to that of 30d. It indicated that PV expression in the adult rat OB was affected irreversibly by deafferentation.6. Effects of ZnSC>4-lesioned olfactory epithelium(OE) on CR expression of adult rat olfactory bulb(OB).10, 20, 30 and 60 days after ZnSO4 treatment, there was no significant diference in CR-IR between control OB and experimental OB. It indicated that CR expression in the adult rat OB was not affected by deafferentation.7. Effects of ZnS04-lesioned olfactory epithelium(OE) on ErbB-3 expression of adult rat olfactory bulb(OB).10, 20, 30d after ZnSO4 treatment, the number of ErbB-3-IR cells of experimental OB decreased gradually, the result of 60d was similar to that of 30d. It indicated that ErbB-3 expression in the adult rat OB was affected irreversibly by deafferentation.8. Effects of Triton X-100-Iesioned olfactory epithelium(OE) on TH, CB and PV expression of adult rat olfactory bulb(OB).30d after Triton X-100 treatment, compared with control, the number of TH-,CB- and PV-IR cells of experimental OB decreased markedly, the number of TH-,CB- and PV-IR cells of 45d were higher than that of 30d, results of 60d were similar to that of the control. It indicated that expression of TH, CB and PV in adult rat OB was regulated reversibly by deafferentation induced by Triton X-100.Conclusion:1. On the whole, the immunohistochemical localization of TH, OMP, CR, PV, CB, ErbB-3, ErbB-4 in the OB of adult Sprague Dawley rat, and OMP, CR, PV, CB, ErbB-3, ErbB-4 and c-fos in the postnatal developmental ICR mice OB were similar to the previous results. Nevertheless, the immunohistochemical localization of ErbB-3 was completely different from the previous reports. The expression of ErbB-3 is demonstrated to be restricted to the ensheathing cells of the olfactory nerve layer (ONL) of adult mice and rats, contrarily, in our study, no ErbB-3 was detected in the ONL of adult rats and postnatal developmental ICR mice OB, we observed manyErbB-3-IR cells in the internal plexiform layer (IPL), granule cell layer (GCL) and subependymal layer (SEL), some in the glomerular layer (GL) and external plexiform layer( EPL). In previous studies, the fluorescent tyramide signal amplification method or immunofluorescence histochem-istry was used, we firstly studied the expression of ErbB-3 in the OB by immunohistochemistry ABC method.2. After OE lesioned by intranasal irrigation with ZnSC>4, TH, CB, PV and ErbB-3 expression in the OB ipsilateral to lesioned naris of adult SD rat OB decreased irreversibly. CR expression was not affected by it.3. After OE lesioned by intranasal irrigation with Triton X-100, TH, CB and PV expression in the OB ipsilateral to lesioned naris of adult SD rat OB showed reversible change.
