Fill The Bone Sustained-release Tablets Pharmaceutical Research

"Buguzhi sustained-release tablet" is a modern Chinese medicinal, which is composed of Psoralen Corylifolia L. and Radix Puerariae. The Chinese medicinal is to treat the primary osteoporosis. This recipe has good curative-effect clinically. The thesis is to screen out the activecomponents from Psoralen Corylifolia L. by treating the osteoblast in vitro and the experiment of estrogen action in rats'vivo. Basing these researches, we study the sustained-release preparationand its quality in vivo and vitro evaluation.I screen out the active compound by the systemic solvent distillation method, sillaca gel chromatrography, and pharmacodynamic experiment in vivo and in vitro. From the active compound I separate 11 constituents, which structures were identified by NMR-1 H , HPLC-MS ,. IR , UV methods and so on. Two coumarin consitituents and three flavone consitituents have been proved to be the active ones because they can speed the growth of the osteoblasts. These researches prove that the coumarin compounds and the flavone compounds are active to treat the primary osteoporosis.I study the distillation and the separation of the active compounds by using the content evaluation of the coumarin and the flavone, and ascertain the best technologic parameters, and get the high pure semi-products. The total content of three batches production from the pilotscale experiments is above 50 percent, which proves that the technologic parameters is reproducible, feasible and suitable to produce in the factory.To optimize extraction and separation techniques of the total flavonoid from Radix Pueraria, I use the content of puerariae and the total flavonoid as the evaluation. The total flavonoid's purity of the three batches pilotscal production is 80%.
