Glucocorticoid inhalation has been used as principal medicine with positive therapeutic efficacy and a little adverse effect . However , there are many patients who have to depend on oral steroids to control condition and the condition will become worse with the oral steroid being stopped to take. The sort of asthma is named as steroid-dependent asthma(SDA) which is belong to difficultly treated asthma .There are continuously existing airway inflammation and severed damaged HPA axle on SDA during the steroid withdrawal. The inflammation is caused by T helper cell subsets of out-of-balance in proportion Th1/Th2, namely the function of Th1 (mainly secreteγ-interferon, IFN-γ) is suppressed, Th2 is in the station of activating, the latter secreting cytokine such as interleukin-4 (IF-4), interleukin-5 (IF-5) which accelerate eosinophil (EOS), mastocyte, lymphocyte,etc. to promote inflammatory cell differentiate, assemble and activate constantly and IgE synthesize,which leads to the fact that the responsivity of airway increases continuously. A great deal of glucocorticoid medicine at the same time regulate competence of duplicating of GCRmRNA backward, make its quantity reduce or the activation become reduced,cause patients' reliance on the glucocorticoid. Glucocorticoid once was reduced or stopped using that can cause the asthma to recur, even cause the death seriously. Therefore we must seek to the effective treatment project that can not only resume the suppressed HPA axle function for the glucocorticoid is used for a long time in a large amount, but also regulate Th1/Th2 ratio, reduce high responsivity of airway, reduce glucocorticoid treat consumption, lighten whole body from bad reaction, is it improve in an all-round way to patient quality to realize.In the opinions of my advisor professor Wu WeiPing, deficiency of yin and yang, stiring up of deficient wind, accumulation of phlegm and blood stasis in the body, simultaneous occurrence of cold and heat syndrome are essential pathogenesis of the steroid-dependent asthma(SDA) after use the glucocorticoid as the leading treatment medication. WuMei pill is the major prescription of Jueyin syndrome in Theory of Cold-Induced Febrile,which can supplement both the vital energy and blood, regulate cold and heat and conform to the pathogenesis of SDA.So it can help to dismantle steroid and control symptom of asthma.1. ObjectiveThe purpose of this experiment is to imitate SDA patients' pathology change,so we inject dexamethasone on the venter of the model mice that have asthma. Through study the asthma model mice that treated by glucocorticoid during the steroid withdrawal and after use traditional medicine and Western medicine the airway inflammation and HPA axle corresponding change situation, announce the evolvement rule and inherent material base for steroid-dependent asthma during the steroid withdrawal, probe into the function mechanism of WuMei pill to treat airway inflammation in asthma and resume HPA axle function.2.Method2.1 The function of lung is detected by the lung function instrument for minitype animal.
