Flavonolignan is a kind of flavones which is coupled by catechol flavones and phenylpropanoids. A lot of researches indicated that flavonolignans have many bioactivies such as antioxidant, hepatoprotective, anti-tumor and lowering blood lipid. The representative compounds of flavonolignans are silybin, isosilybin, silichristin, silidianin and 2, 3-dehydrosilybin which were isolated from silymarin. Silybin has the most activity among them. Some of the compounds have been applied in the clinical test, but the bioavailability is limited due to their low water solubility. It is meaningful to investigate the structure-activity relationships of flavonolignans. The E ring analogues of silybin were synthesized by a new method, while the hemi-transformation of silybin on its B ring, E ring and 3, 23, 7, 20 positions was also performed. Some of the compounds' antioxidant activities were investigated.Dihydroflavonol is another kind of flavone in which 2,3-position is a single bond and 3-position is substituted by hydroxyl. It was found to possess a lot of biological activities such as antioxidant, antitumor and hepatoprotective. Many dihydroflavonols such as taxifolin, astilbin, ampelopsin show excellent activities. But the researches about the SARs of dihydroflavonol are still rarely. To investigate its relationships, we synthesized 5,6,7-trioxygenated dihydroflavonols and modified their 7-position and B ring. The scavenging free radical activities of some compounds were tested.Sinapyl alcohol derivatives are the derivatives of 3,5-dimethoxy-4-hydroxycinnanic alcohol. Researches indicated that sinapyl alcohol derivatives isolated from natural products showed excellent cell cytotoxicity. It supplied an excellent lead compound for anti-tumor. To investigate their SARs, a more convenient and efficient synthesis route of sinapyl alcohol was designed and a series of derivaties were also prepared.In summary, 69 target molecules and 104 intermediates were synthesized in which 123 are new compounds.
