Lasianthus acuminatissimus Merr. is a plant of the Rubiaceae family, distributed in the southern provinces of China. It is used in traditional Chinese folk medicine for the treatment of rheumatoid arthritis. However, the investigations on the chemical constituents of plant have been few.In a search for the active constituents from the plant, the ethanol extract of the roots was subjected to a systematical chemical investigation, leading to the isolation of thirty-five compounds by solvent and chromatography methods, and thirty structures (Figure 1) were elucidated on the basis of physical and chemical evidence and modern spectral analysis (including IR,UV,1H-NMR,13C-NMR,HMQC,HMBC,DEPT,EI-MS,FAB-MS and HR-MS, et al.). Which are: lasianthuoside A, lasianthuoside B, lasianthuoside C, lasianthurin A, lasianthuoactone A, damnacanthol-11-methyl ether, damnacanthol, damnacanthol-3-O-β-D-primeveroside, tricontanoic acid,βconfiguration of mudanoside-A, isoscopletin, nonadecyl alcohol, cis-13-docosenoic acid, 2, 5-dimethoxy-1, 4-benzoquinone, asperulosidic acid, asperuloside, codonolactone, isoscopletin-6-O-β-D-glucopyranoside, (-)-pinoresinol 4-O-β-D-glucopyranoside, uncargenin C, tetracosanoic acid, deacetyl asperulosidic acid, non-glycosidic iridoid, 2, 6-dimethoxy-4-hydroxyphenol-1-O-β-D- glucopyranoside, tachioside, isotachioside, scandoside, benzoic acid,β-sitosterol, daucosterol. Among them, lasianthuoside B, lasianthuoside C, lasianthurin A, lasianthuoactone A are five new compounds, seventeen compounds are obtained from the family for the first time, all compounds were reported from this plant for the first time.In addition to a preliminary evaluation in treating rheumatoid arthritis by the AA animal model was carried out, two iridoid glycosides (asperuloside and deacetyl asperulosidic acid) were found to be the bioactive compounds in the plant. The antitumor activity of seven anthraquinones against human 2780 cells was also evaluated, and three new anthraquinone glycosides showed significant bioactivity. And compound 2, 5-dimethoxy-1, 4- benzoquinone has a significant inhibition on release ofβ-glucuronidase rat polymorphous nuclear leukocytes activated by platelet activating factor (PAF), which revealed it was an active anti-inflammation compound in the plant.
