1 Objective : By the pathology change of bone erosion of joint and change of RANKL, OPG, TUNKL by in situ hybridization after cure, We can learn the meachanism of bone erosion and effect using medicine.Rheumatoid arthritis is a course of high bone erosion. The evdivent coming from experiment, It is the need of bone erosion by Osteoblast and Osteoclast. Osteoclast activation need the action of RANKL and it receptor-RANK.As one of the nember of tumor necrosis factor (TNF), OPG can joint the RANKL by competitive combination .and stop the joint to RANK. It can inhibit the activation of Osteoclast and incress the apoptosis of Osteoclast to inhibit the absole of bone.The erosion of bone issue and cartilage come from active of synovialmembrane, because the shorge apoptosis of synovial membrane cell.Methods: Based on the collagen induced arthritis, Each group give commondose of Chinease herbal medicine (Yubitang) , high dose of Chinease herbalmedicine (Yubitang) or Methotrexate(MTX) to cure. In the Zhenghou of RA,The Yubitang is suit of Ganshenkuixu.Methods1 Experimental study1.1 Group dispensation50 health SD famale mice were divided into 5 groups (normal control,model group, common dose group , high dose group and MTX group) in randomeach group contains 10 mice.1. 2 Model making process and methods of experiment①Model group:10 mise were immunized with the mixture of 0.1ml FCA and250 ug CII which was injected into ankle joint skin of right feet. They are' t have medine to cure, Every day give them 0.9%NS 2ml.(2) Normal control: treated with nothing.(3) common Chinease herb group:10 mice were treated as the model group except that they were fed with 2ml Chinease herb every after seventh day of making model.? high Chinease herb group: treated as common Chinease herb group except the concentration of medince as two double as them. (§) MTX group: treated as common Chinease herb group except give them MTX 0. lmg/2ml every week.At the end of experiment , Each of groups mice both ankle joints were cut off for pathological examination., and 4~6ml blood was taken from artery of abdomen. 1.3 Items observed ?General detection: mice weights thickness of feet.(2) Pathological detection: HE staining of joint' s cartilage(3) In situ hybridization detection: 0PG^ RANKLResults2.1 Mice weightMice weight of Chinease herb group, MTX group, Model group have significantly decreased compareing with Normal control from 14 days, but Mice weight of Chinease herb group, MTX group, Model group have significantly, weight of Model group are light, and Chinease herb group, MTX group, Normal control have' nt significantly difficult from twenty-one day.2.2 The swollen of mice feetThe swollen of mice feet of Chinease herb group, MTX group , Model group come after making modlehave significantly difficult compareing with Normal control.In twenty-one day, The swollen of mice feet are highest. But The swollen of mice feet of Chinease herb group, MTX group are decress .similar with Normal control about twenty-eight day. Compareing with them ,The swollen of mice feet of Model group do not significantly decress.2.3 Mean Optical Density of OPGAt the end of trial the mean Optical Density about OPG of Chinease herb group, MTX group are much larger than Model group, there is a significant difference between them, while there are no significant difference among these three groups of cure group, and between Normal control. Among them , high Chinease herb group is the largest.2.4 Positive cartilage cells of OPGThere are more OPG positive cartilage cells in joints cartilage of Chinease herb group, MTX group than normal control and model group,. have significant difference .Normal control and model group are simile .Among the cure group , high Chinease herb group is the largest, common Chinease herb group is the larger, they are have significant difference.2.5 Mean Optical Density of RANKLAt the end of trial the mean Optical Density about RANKL of Chinease herb group, MTX group, Model group are incress compressing with Normal control,there is a significant difference between them, while there are no significant difference among these three groups of cure model group, andbetween with Model group.2.6 Positive cartilage cells of RANKLAt the end of trial the Positive cartilage cells of RANKL of Chinease herb group, MTX group, Model group are incress compressing with Normal control, there is a significant difference between them, while there are no significant difference among these three groups of cure model group ,and between with Model group.2.7 The apoptosis rate cartilage cellThere isn' t significant difference among each group.2.8 The apoptosis rate synovial membrane cellThere are significant difference between Chinease herb group, MTX group, Normal group and Model group. Among them .Significant difference between high Chinease herb group, MTX group, Model group and common Chinease herb group. High Chinease herb group, MTX group, Normal control are simile.3 Pathological results 3.1 Normal control :The structure of bone and cartilage of joins are intact without infiltration of inflammatory cells in synovial membrane and synovial membrane over-growth. 3. 2 Model groupSynovial membrane of joints over grown with infiltration of inflammatory cells and several lays of synovial cells aligned in disorder. Pannus formation .cartilage degradation and fibrosis. 3.3 Cure groupThe syndromes appeared in Model group were reduced significantly.4 Conclusion:The OPG of Chinease herb group, MTX group are much larger than Model group, there is a significant difference between them, OPG can joint the RANKL by competitive combination and stop the joint to RANK. It can inhibit the activation of Osteoclast to decress the damage of cartilage. While high Chinease herb group is the best, among cure group.There are significant difference between Chinease herb group, MTX group, Normal control and Model group. It show that the erosion of bone issue and cartilage are decress by incressing apoptosis of synovial membrane cell.There are not significant difference between Chinease herb group, MTX group, Normal control and Model. Perhaps 1. The time of making model is short , the damage of erosion are not very appear. 2. When damage of erosion are early, the cartilage can repair by themselves.The Chinease herbal medicine of YU BI TANG can incress the rate of OPG/RANKL and apoptosis of synovial membrane cell to inhibit the damage of cartilage.
