Part IObjectives To establish a stable primary hepatocellular carcinoma(HCC) model of C57BL/6J mice induced by diethylnitrosamines (DEN), Carbon Tetrachloride ( CCl4) and ethanol. Methods 90 C57BL/6J mice were induced for twenty weeks by DEN/CCl4 ethanol , and then the occurrence and development of HCC were observed, and another 10 C57BL/6J mice were used as normal control(no drug was given). RT-PCR method was used to detect murine fetoprotein(mAFP) mRNA expression in tumor tissues. Results 71 of 90 mice had been induced into HCC(78.89%), cirrhosis had been observed among 11 of 90 mice( 12.22%), and 8 of them died of poisonous hepatitis and necrosis at earlier period. There were high expression of AFP mRNA in cancer tissue and no pathologic changes were observed for liver tissues of the control group. The histological pathologic changes of HCC were classified high or middle differentiation. Conclusions HCC with secretion of murine alpha fetoprotein model of C57BL/6J mice induced by DEN/CCl4/ethanol was successfully established . The model had a stable carcinogenesis for HCC. It had experienced a course similar to human HCC development.
