We investigated the effect of KW-2228 (rhG-CSF) on bone marrow suppression caused by high-dose and multicycle administration of Mitomycin C (MMC) in C3H/HeJ JcL mice. MMC was injected intraperitoneally at the doses of 6 or 7mg/Kg. KW-2228 (rhG-CSF) was administered subcutaneously 24 hours after MMC-inject ion (at the dose of 5ug/mouse/ day) for 14 times. We determined 21 days as one cycle, and in the experiments 2 cycles have been chosen. We found that KW-2228 could shorten the period of the MMC-induced neutrocytopenia and accelarate the generation of neutrophils. Further more,KW-2228 also improve the drug-induced reduction of platelets and lymphocytes in all living mice administered with MMC at the dose of 6mg/Kg for 2 cycles or 7mg/Kg. As for the body weight, there was no difference between MMC used alone and combination of MMC and KW-2228. So was the mortality of mice treated between with the MMC (7mg/Kg) alone and with combination of MMC and KW-2228. Therefore, KW-2228 administered simultaneously in combined with chemotherapeutic drugs could accelarate the regeneration of neutrophils at early stage, and could also enhance the recovery of platelets and lymphocytes. Meanwhile, these effects of KW-2228 did not improve the mortality and the body weight loss of the mice treated with MMC.
