The pharmacokinetics and pharmacodynamics of pancuronium in acute moderate isovolemic hemodilution were studied in thirty ASA I patients who were undergoing plastic surger operation.Following pethidine and droperidot premedi-cation, anesthesia was induced with fentanyl(2ug/ kg), thiopentone(4-6mg/kg). To facilitate endo-tracheal intubation, 2% lidocaine was applied topically to the larynx without using any relaxant. Anesthesia was maintained with 60% nitrous oxide in oxygen. Before operation, the venous blood was withdrawn and replacement with 6% dextran(molecular weigh 70,000) and Ringer'slactate solution. The Hct decreased from pre—hemodilution 41% to after hemodilution 28.02%. the hemoglobin from 14. 5% to 10.1l%(P<0.05). Neuromuscular function was monitored by means of accelerometry, T0F stimulation was applied. In the first part of the study, the meuro— muscular blocking potency of pancuronium was determined with cumulative dose-response tech(?)ique. After a bolus injection of pancuronium(ED50), the peak effect(%de-prdssion) and onset of time were recorded, after peak effect was achieved another dose equal to 2ED95 of pancuronium was injected and duration was observed. The results...
