BackgroundRat Glutathione S-transferases P (GST-P) are identified to be closely related to cellular transformation processes, chemical carcinogenesis and tumor cell multidrug resistance. Rat GST-P is hardly expressed in rat normal liver cells, but it is specially expressed in precancerous lesions and in hepatoma induced by carcinogens or spontaneously arising hepatomas. On account of this, GST-P is regarded as one of the most stable and reliable preneoplastic and neoplastic marker enzymes. Inquiry into the expression of rat GST-P gene revealed that the induced expression of GST-P gene during chemically carcinogenic processes was shown to be regulated mainly at the transcriptional level. Therefore clarifying the transcriptional regulatory mechanism of GST-P gene appears to be very important to elucidate the induced expression of GST-P gene during carcinogenic processes and to determine the mechanisms of chemical carcinogenesis, which will provide the theory for the therapy and precaution to carcinoma.ObjectionTo investigate the diversity of the mechanisms of rat GST-P gene expression regulation and the relation between rat GST-P gene expression and carcinogenesis. To search the trans-action factors binding to the enhancer element of rat GST-P (GPEI).Methods and results1. To study the effect and mechanism of chemicals on rat GST-P gene expression Investigate the mechanism of the induction of rat GST-P gene expression by...
