Objective:1 The aim of this study is to establish the method for isolation and in vitro culture expansion of adult human bone marrow-derived mesenchymal stem cells (MSCs) and to investigate their biological characteristics.2 To explore the beneficial effect of MSC on in vitro expansion of cord blood (CB) hematopoietic stem cells (HSCs) and adhesion molecules expression of CB CD34~+ cells.3 To study the suppressive effect of MSCs on immune function. Methods:1 MSCs were isolated based on their selective adherence, compared to haematopoietic cells, to plastic surfaces. Monouclear cells(MNCs) or whole marrow cells were obtained from normal human bone marrow aspirates and cultured in Dulbecco's Modified Eagle's /F12 Medium containing 10% fetal bovine serum. Their differentiation function was investigated with specific induction culture system. The FACS was performed to examine the expression of cell surface molecules and DNA content.2 CB CD34~+cells were expanded on MSC layer or MSC-free culture systems and the effect of MSC on expansion potential and expression of adherent molecules on CB CD34~+cells were investigated.
