Background Chemotherapy resistance is one of most major factors to cause failure in treating carcinoma and leukaemia. The mechanism of Chemotherapy resistance in cancer cells is complex which include many aspects. Besides drug pumped out of cell, enhancing activity of detoxication enzyme and strengthening the function of DNA repair, anti-apoptosis is also an key mechanism in that issue. multidrug-resistance protein, P-gp and MRP, multidrug-resistance protein, are members of ABC family. P-gp and MRP are component of ATP-dependent protein channel on membrane of cell which could pump drugs out of cells and protect cells to be hurt by drugs, but when the power supply is blocked from ATP to P-gp and MRP it can not restore the sensitive to chemotherapy in cancer cells. Further study showed that P-gp and MRP could inhibit the activity of caspase-8 so that they could block the pathway of caspase- dependent apoptosis. For the moment most study related to convert multidrug-resistance is focus on positioning the sites of those mechanism mentioned above. Whereas for the toxic side effects or the mechanism is still unknowable, there is still no effective way to overcome the chemotherapy resistance clinically.Chloramphenicol (CAP) is a small molecule substance with a character of anti bacterial being used in clinic.
