PART ONECONSTRUCTION OF VECTOR CARRYING HUMAN INSULIN-LIKE GROWTH FACTOR-1 GENE AND THE GENE EXPRESSION IN EUCARYOTEInsulin-like growth factor-1(IGF-1) is one of the most important regulate factors which can stimulate cartilage cell to differentiate and synthesis collagen typeâ…¡and proteoglycan to maintain cartilage phaenotype. With the development of gene transfection research, it is completely possible to introduct exogenous gene to target cells and get it expressed with high efficience and to affect cells in some aspects. Here, we constructed vector contained hIGF-1 and the marker gene , enhanced-green fluorescent protein gene, and identified the vector construction, which followed by determination of the gene expression in the eukaryocyte, HEK293 cells. We showed that the recombinated vector may be a great convenient tool for gene modify of mesenchymal stem cells, the preferred seed cells for cartilage tissue engineering. Objective To construct a eukaryotic vectors carrying truncated human insulin-like growth factor-1 gene and marker gene, enhanced-green...
