The blood vascular system has a continuous lining which consists of a single layer of endothelial cells. The intact functional endothelial cells play important roles in maintaining the self-stabilization of vasculature and normal blood circulation. The abnormal expression of TF gene may directly affect the anti-thrombotic properties of endothelial cells. The thrombosis initiated by TF plays a role in the occurrence and development of thrombotic disease. Recently, the mechanism of endothelial TF gene expression and regulation induced by TNF α or Ang II was paid close attention, but few researches are carried out. The effect of 7643, an effective component of Danshen, on TF gene expression have not been reported yet. The aim of this study is to explore the effect of TNF a or Ang II on the function of HUVECs and ECV304 cells including protein activity, mRNA and transcriptional regulation respectively. The intervenient effect of 7643 was also observed. The following aspects are studied:1) the effect of TNF α or Ang II on TF activity of the endothelial cell surface;2) the effect of TNF α or Ang II on TF mRNA expression in vascular endothelial cells;3) the regulatory effect of upstream sequence on TNF α or Ang II induced TF gene transcription in vascular endothelial cells and the intervenient effect of 7643;4) the effect of TNF α or Ang II on the expression of NF- κ B in vascular endothelial cells;5) the effect of TNF α or Ang II on the activation of NF- κ B in vascular endothelial cells;6) the role of calcium ion (Ca2+) in upstream sequence controlling TNF α or Ang II -...
